桑树Aux/IAA基因家族鉴定与IBA处理下表达模式分析 被引量:4

摘要 【目的】在全基因组范围内鉴定桑树Aux/IAA基因家族成员,并研究在IBA处理下对该家族基因表达模式的影响,为深入研究桑树Aux/IAA基因家族的功能提供依据。【方法】通过本地川桑全基因组数据对桑树Aux/IAA基因家族成员进行鉴定分析,包括蛋白质理化性质分析、系统进化树构建、基因结构分析、蛋白质三级结构及蛋白互作关联分析、启动子顺式作用元件分析。并以“强桑1号”扦插枝为试验材料,对其进行1000 mg·L^(-1) IBA处理和清水对照处理,分析处理后10、20、30、40 d的MnAux/IAA基因家族的表达模式。【结果】MnAux/IAA基因家族共有51个成员,分为8个亚家族,AtIAA基因家族分布在其中6个亚家族中;51个基因家族成员均有外显子和内含子,仅有1个成员无UTR;顺式作用元件表明,MnAux/IAA基因家族对光、激素、逆境胁迫等都有相应的调控作用;蛋白质同族进化序列的基因结构相似度较高,但族间差异较大;IBA处理后,47个基因表达量会有不同程度的提升,而基因MnAux/IAA34、MnAux/IAA33、MnAux/IAA32在表达量上有较大程度的下降。【结论】MnAux/IAA基因家族功能相对稳定,在家族进化过程中无明显变化;IBA具有促进Aux/IAA基因表达的作用;MnAux/IAA34、MnAux/IAA33、MnAux/IAA32与AtARF2同源性较高,推测具有调控叶片衰老的作用。 【Objective】To identify Aux/IAA gene family members in the whole genome,and study the effects of IBA treatment on the gene expression pattern of the family,so as to provide a basis for further research on the function of Aux/IAA gene family in Morus notabilis.【Method】The Aux/IAA gene family members of Morus notabilis were identified and analyzed through local Morus notabilis whole genome data,including protein physicochemical property analysis,phylogenetic tree construction,gene structure analysis,protein tertiary structure and protein interaction association analysis,and promoter cis-acting element analysis.The cuttings of“Qiangsang No.1”were treated with 1000 mg·L^(-1) IBA and water control,and the expression patterns of MnAux/IAA gene family on the 10th,20th,30th and 40th day repectively after treatment were analyzed.【Result】The MnAux/IAA gene family has 51 members,which are divided into 8 subfamilies,and the AtIAA gene family is distributed in 6 subfamilies;All 51 gene family members had exons and introns,and only 1 member had inexistence UTR;The cis-acting elements indicated that the MnAux/IAA gene family had corresponding regulatory effects on light,hormones and stress;The similarity of gene structure of protein isogenetic sequences is high,but there exists greater difference between families;After IBA treatment,the expression levels of most genes were increased to varying degrees,and the expression levels of genes(MnAux/IAA34,MnAux/IAA33,MnAux/IAA32)were decreased to a large extent.【Conclusion】The function of the MnAux/IAA gene family is relatively stable,and there is no significant change in the family evolution;IBA can promote the expression of Aux/IAA gene;MnAux/IAA34,MnAux/IAA33,MnAux/IAA32 have high homology with AtARF2,which suggests that they may play a role in regulating leaf senescence.
