沙棘果渣黄酮微波提取工艺优化及其纯化研究 被引量:4

Study on Microwave Extraction Technology Optimization and Purification of Flavonoids from Fruit Residue of Hippophae rhamnoides

摘要 采用单因素试验研究乙醇体积分数、料液比、微波时间和微波功率对沙棘果渣中黄酮提取量的影响,在单因素试验的基础上采用响应面优化沙棘黄酮提取工艺,并利用大孔树脂对沙棘黄酮进行纯化。结果表明,乙醇体积分数70%、料液比1︰25 (g/mL)、微波时间2.5 min时,沙棘黄酮提取量达22.13 mg/g。在大孔树脂AB-8纯化沙棘黄酮研究中,上样浓度2 mg/mL、上样流速1.5 mL/min、上样量70 mL、洗脱液流速2 mL/min时,沙棘果渣提取物中总黄酮含量从17.3%提高到31.50%。试验结果为沙棘副产物的综合利用和新产品开发提供参考。 The single factor experiment was used to explore the effects of ethanol concentration,ratio of material to liquid,microwave time,and microwave power on the extraction content of flavonoids from fruit residue of Hippophae rhamnoides.On the basis of single factor experiment,response surface methodology was optimized by extraction technology of flavonoids from Hippophae rhamnoides,and macroporous resin was used to purify flavonoids from Hippophae rhamnoides.The results showed that when the ethanol concentration was 70%,the ratio of material to liquid was 1︰25(g/mL),and the microwave time was 2.5 min,the extraction amount of flavonoid from Hippophae rhamnoides was as high as 22.13 mg/g.The purification of flavonoids from Hippophae rhamnoides,using macroporous resin AB-8,when the concentration of loading solution was 2mg/m L,the flow rate of loading sample was 1.5 mL/min,the loading volume was 70 mL,and the elution flow rate was 2 mL/min,the total flavonoids content form fruit residue of Hippophae rhamnoides extract increased from 17.3%to 31.50%.The research results can provide some reference for the comprehensive utilization of fruit residue of Hippophae rhamnoides.
