摘要
目的:探讨黄芪建中汤对脑缺血小鼠的抗疲劳作用及初步作用机制。方法:将40只昆明种小鼠随机分成5组:对照组、脑缺血模型组、黄芪建中汤低剂量组、黄芪建中汤中剂量组、黄芪建中汤高剂量组,每组8只。造模术后第1天开始给药,黄芪建中汤低、中、高剂量组分别给予生药量为0.154g/ml、0.308g/ml、0.616g/ml水煎剂0.5ml/d灌胃,对照组与脑缺血模型组则给予等量生理盐水,给药14d后,观察各组小鼠负重游泳力竭时间、在疲劳转棒仪上停留时间,同时测定小鼠四肢肌力及空腹血糖。结果:与脑缺血模型组比较,给药组小鼠负重游泳时间均明显延长,黄芪建中汤低、高剂量组小鼠在疲劳转棒仪上的停留时间增加,不同剂量给药组小鼠四肢肌力均显著增强(P<0.05),但各组小鼠空腹血糖水平无显著性差异(P>0.05)。结论:黄芪建中汤对小鼠脑缺血后疲劳具有较好的改善作用,特别是黄芪建中汤高剂量组,但其抗疲劳作用机制还需更加深入的探索。
Objective:To explore the anti-fatigue effects and its preliminary mechanism of Huangqi Jianzhong decoction on mice with cerebral ischemia.Methods:40 Kunming mice were randomly divided into 5 groups:control group,cerebral ischemia model group,Huangqi Jianzhong decoction low dose group,Huangqi Jianzhong decoction medium dose group and Huangqi Jianzhong decoction high dose group,with 8 mice in each group.The administration was started on the first day after modeling,the crude drug dosage of 0.154g/ml,0.308g/ml and 0.616g/ml Huangqi Jianzhong decoction were given 0.5ml/d in the low,medium and high dose groups.And the control group and the cerebral ischemia model group were given the same amount of normal saline each time.After 14 days of administration,the exhaustion time of weight-bearing swimming,the residence time of mice on fatigue rotors,the four limbs’muscular strength of mice and fasting blood glucose levels were also measured.Results:Compared with the cerebral ischemia model group,the exhaustion time of weight-bearing swimming of the mice in Huangqi Jianzhong decoction group was significantly prolonged,the residence time of the mice on the fatigue rotors in the low and high dose groups was increased,and the four limbs’muscular strength were also significantly increased(P<0.05).But there was no significant difference in fasting blood glucose levels among five groups(P>0.05).Conclusion:Huangqi Jianzhong decoction has a good effect on fatigue after cerebral ischemia in mice,especially in Huangqi Jianzhong decoction high dose group,but its mechanism still needs further exploration.
