摘要
为了探寻参苓白术散药味增减对葡聚糖硫酸钠诱导的溃疡性结肠炎小鼠的治疗效果,试验将30只雌性C57BL/6小鼠(6~8周龄,体重为18~20 g)随机分成5组,分别为空白对照组、模型组及高、中、低剂量组,每组6只。第1周各组适应性饲养;第2周除空白对照组外,每组用3%葡聚糖硫酸钠水溶液自由饮水,连用7 d,建立急性溃疡性结肠炎模型;第3周更换正常饮水,空白对照组和模型组灌胃生理盐水,高、中、低剂量组按体重分别灌胃0.2,0.1,0.05 mL/10 g中药药液。观察并记录小鼠临床症状,采集全血分离血清,检测血清中免疫球蛋白G(IgG)、免疫球蛋白A(IgA)、白细胞介素-8(IL-8)、白细胞介素-10(IL-10)、二胺氧化酶(DAO)、D-乳酸的质量浓度,同时制作小鼠结肠组织病理切片。结果表明:与空白对照组比较,模型组血清中IL-8、IgA、IgG、DAO、D-乳酸的质量浓度极显著升高(P<0.01),IL-10的质量浓度极显著降低(P<0.01);给药后,高、中、低剂量组小鼠的炎症均有不同程度缓解,IL-8、IgA、IgG、DAO、D-乳酸的质量浓度均有不同程度下降,IL-10的质量浓度上升。说明参苓白术散药味增减对葡聚糖硫酸钠诱导的溃疡性结肠炎有良好的治疗作用,在本试验条件下,按体重0.2 mL/10 g给药治疗效果最好。
In order to explore the therapeutic effects of Shenling Baizhu powder on ulcerative colitis in mice induced by dextran sulfate sodium,30 female C57BL/6 mice(6-8 weeks old,weighing 18-20 g)were randomly divided into five groups,namely blank control group,model group,and high,medium,and low dose groups,with six mice in each group.In the first week,each group underwent adaptive feeding.In the second week,except for the blank control group,each group was given 3%dextran sulfate sodium aqueous solution for free drinking water for seven days to establish an acute ulcerative colitis model.In the third week,normal drinking water was replaced,and the blank control group and model group were given physiological saline by gavage.The high,medium,and low dose groups of the formula were given 0.2,0.1,0.05 mL/10 g of traditional Chinese medicine solution by gavage according to their body weight.Clinical symptoms of mice were observed and recorded;serum was collected from whole blood;mass concentration changes of immunoglobulin G(IgG),immunoglobulin A(IgA),interleukin-8(IL-8),interleukin-10(IL-10),diamine oxidase(DA0)and D-lactic acid in serum were detected,and pathological sections of colon were made.The results showed that compared with the blank control group,the mass concentrations of IL-8,IgA,IgG,DAO,D-lactate in the serum of the model group were significantly increased(P<0.01),while the mass concentration of IL-10 was significantly reduced(P<0.01).After administration,the high,medium,and low dose groups of the mice was alleviated to various degrees,and the mass concentrations of IL-8,IgA,IgG,DAO,D-lactate were all reduced to various degrees,while the mass concentration of IL-10 increased.These results indicated that Shenling Baizhu powder had a good therapeutic effect on ulcerative colitis induced by dextran sulfate sodium.Under the conditions of this experiment,the best therapeutic effect was given at O0.2 mL/10 g of body weight.
