摘要
为培育灵芝优良新品种,对其进行太空诱变育种,筛选出15株诱变菌株。对它们的活性成分含量进行测定,结果显示,诱变菌株T6、T12、T15的多糖含量和T10、T15、T19的三萜及甾醇含量均显著高于出发菌株T0,T35的多酚含量显著高于出发菌株T0。体外抗氧化活性评价结果显示,诱变菌株T10、T19、T35醇提物的总还原力显著高于出发菌株T0,但是参试菌株水提物的总还原力与出发菌株T0没有显著差异;诱变菌株T15水提物的DPPH自由基清除率和醇提物的超氧阴离子自由基清除率以及T10、T19、T35的醇提物的DPPH自由基清除率和水提物的超氧阴离子自由基清除率均显著高于出发菌株。这些结果表明航天诱变育种能够改变药用真菌的活性成分含量和抗氧化能力,通过试验筛选可以获得活性成分含量高和抗氧化作用强的优良菌株,是优良新品种培育的有效途径之一。该试验结果可为微生物航天诱变育种研究提供参考,也有望为灵芝工业化生产提供更优良的菌种。
In order to cultivate excellent new varieties of Ganoderma lucidum,space mutation breeding was conducted and 15 mutagenic strains were screened.The content of their active ingredients was measured,and the results showed that the polysaccharide content of mutagenic strains T6,T12,and T15,as well as the triterpenoid and sterol content of T10,T15,and T19,were significantly higher than those of the original strain TO;The polyphenol content of T35 was significantly higher than that of the original strain To.The in vitro antioxidant activity evaluation results showed that the total reducing power of the alcohol extracts of mutagenic strains T10,T19,and T35 was significantly higher than that of the original strain To,but there was no significant difference in the total reducing power of the water extracts of the test strains compared to the original strain TO;The DPPH free radical scavenging rate of the water extract and the superoxide anion free radical scavenging rate of the alcohol extract of mutagenic strain T15 were significantly higher than those of the original strain;The DPPH free radical scavenging rate of the alcohol extract and the superoxide anion free radical scavenging rate of the water extract of mutagenic strains T10,T19,and T35 were significantly higher than those of the original strain T0.These results indicate that space mutation breeding can change the active ingredient content and antioxidant ability of medicinal fungi.Through experimental screening,excellent strains with high active ingredient content and strong antioxidant activity can be obtained,which is one of the effective ways to cultivate excellent new varieties.This experiment provides a reference for the research of microbial space mutation breeding,and is also expected to provide better strains for the industrial production of G.lucidum.
