胆固醇代谢在结直肠癌调控中的作用研究进展

摘要 胆固醇代谢是近年癌症治疗探索的热点,阐述其复杂的作用机制潜在地奠定了脂-癌相关的分子基础。但目前多数研究仅聚焦于将胆固醇作为产物来理解,并分析其与癌症进展的关系,忽略了其代谢过程中产生的相关衍生物、调节蛋白及免疫细胞的阶段性影响,因此,能否从胆固醇代谢途径中寻找调控结直肠癌(CRC)的靶点成为目前研究的重点。本文从细胞内异常胆固醇代谢途径入手,综述其相关分子机制及信号通路在CRC代谢重编程中的作用,以期为靶向胆固醇治疗CRC提供新思路。 Cholesterol metabolism is a hot topic in exploration of cancer treatment in recent years,and its complex mechanism of action potentially lays a molecular foundation related to lipid-cancer.However,most studies focus only on cholesterol as a product to understand and analyze the progress of cancer,ignoring the phased effects of related derivatives,regulatory proteins and immune cells in its metabolic process,so whether to find targets to regulate colorectal cancer(CRC)from the cholesterol metabolic pathway has become a research focus.This paper reviews the molecular mechanism and the role of signaling pathways in metabolic reprogramming of CRC starting from the abnormal intracellular cholesterol metabolic pathway in order to provide new ideas for the targeted cholesterol therapy of CRC.
