Sirtuins基因家族及其剪接体在小鼠性腺的时空表达谱研究

Study on spatiotemporal expression profiles of Sirtuins gene family and their splices in mouse gonads

摘要 为了研究Sirtuins基因家族候选剪接体在1~8周龄雄性和雌性小鼠性腺的时空表达模式,试验采用半定量PCR方法评估了不同周龄雄性和雌性小鼠性腺Sirt1、Sirt2、Sirt5、Sirt6和Sirt7基因候选剪接体的表达水平。结果表明:性腺Sirt1、Sirt2、Sirt6基因的不同剪接体呈现不同的时空表达谱。卵巢和睾丸Sirt1.1、Sirt1.2基因在6周龄相对表达量极显著低于4周龄(P<0.01),卵巢Sirt1.2基因和睾丸Sirt1.1、Sirt1.2基因在8周龄显著高于6周龄(P<0.05)。卵巢和睾丸Sirt2.1基因相对表达量在4周龄显著或者极显著低于2,6,8周龄(P<0.05或P<0.01)。卵巢Sirt2.2基因相对表达量在4周龄显著高于2周龄且显著低于6周龄(P<0.05),8周龄显著高于6周龄(P<0.05);睾丸Sirt2.2基因相对表达量在8周龄时极显著高于2周龄(P<0.01),显著高于4,6周龄(P<0.05)。睾丸Sirt2.3基因相对表达量在4周龄时显著低于2,6,8周龄(P<0.05)。卵巢Sirt2.4基因相对表达量在4周龄显著高于2周龄(P<0.05),显著低于6,8周龄(P<0.05);睾丸Sirt2.4基因相对表达量在4周龄极显著低于2,6,8周龄(P<0.01),而6周龄极显著高于8周龄(P<0.01)。卵巢Sirt5基因相对表达量6,8周龄显著高于2,4周龄(P<0.05),睾丸Sirt5基因相对表达量在各周龄间差异不显著(P>0.05)。卵巢Sirt6.1基因相对表达量在2周龄极显著高于4,6,8周龄(P<0.01);卵巢Sirt6.2基因相对表达量在2周龄极显著高于4,6周龄(P<0.01),显著高于8周龄(P<0.05),8周龄极显著高于4,6周龄(P<0.01)。睾丸Sirt6.1、Sirt6.2基因相对表达量的趋势相近,均是在2,4,6周龄逐渐下降,8周龄极显著升高(P<0.01)。卵巢Sirt7.1基因相对表达量随周龄增长而降低,睾丸Sirt7.1基因相对表达量在2,4周龄显著高于6,8周龄(P<0.05)。说明Sirt1、Sirt2、Sirt6及其剪接体在小鼠性腺存在不同的时空表达谱,这些不同剪接体在雌雄小鼠性腺发育各阶段可能发挥不同的功能。 In order to investigate the temporal and spatial expression patterns of candidate splicosomes from the Sirtuins gene family in the gonads of male and female mice aged 1 to 8 weeks,semi-quantitative PCR was employed to assess the expression levels of Sirt1,Sirt2,Sirt5,Sirt6,and Sirt7 candidate splicosomes in the gonads of male and female mice at different developmental weeks of age.The results showed that different spliceosomes of Sirt1,Sirt2 and Sirt6 in gonads showed different spatiotemporal expression profiles.The relative expression levels of Sirt1.1 and Sirt1.2 in the ovaries and testes were significantly lower at 6 weeks than thase at 4 weeks(P<0.01).The Sirt1.2 genes in ovary and Sirt1.1 and Sirt1.2 genes in testis were significantly higher at 8 weeks of age than thase at 6 weeks of age(P<0.05).The relative expression of Sirt2.1 gene in ovary and testis at 4 weeks of age was significantly lower than that at 2,6,and 8 weeks of age(P<0.05 or P<0.01).The relative expression levels of Sirt2.2 in the ovaries were significantly higher at 4 weeks of age than thase at 2 and significantly lower than at 6 weeks of age(P<0.05),the relative expression levels of Sirt2.2 in the testes were significantly higher at 8 weeks of age than at 6 weeks of age(P<0.01),and the relative expression levels of Sirt2.3 in the testes were significantly lower at 4 weeks of age than thase at 2 and significantly lower than thase at 6 weeks of age.The relative expression level of Sirt2.4 in the ovaries was significantly higher at 4 weeks of age than that at 2 weeks of age and significantly lower than that at 6,8 weeks of age(P<0.05),The relative expression of Sirt2.4 gene in testis at 4 weeks of age was significantly lower than that at 2,6,8 weeks of age(P<0.01),and significantly higher than that at 6 weeks of age(P<0.01).The relative expression level of Sirt5 gene in ovary at 6 and 8 weeks of age was significantly higher than that at 2 and 4 weeks of age(P<0.05),but there was no significant difference in the relative expression level of Sirt5 gene in testis at all weeks of age(P>0.05).The relative expression level of Sirt6.1 in the ovaries was extermely significantly higher at 2 weeks of age than that at 4,6,8 weeks(P<0.01).The relative expression level of Sirt6.2 in the ovaries was extermely significantly higher at 2 weeks of age than that at 4,6 weeks of age(P<0.01),and significantly higher at 8 weeks of age(P<0.05),and extermely significantly higher at 8 weeks of age than that at 4,6 weeks of age(P<0.01).The relative expression levels of Sirt6.1 and Sirt6.2 genes in testis showed similar trends,gradually decreasing at 2,4,and 6 weeks of age,and significantly or extremely significantly increasing at 8 weeks of age(P<0.05 or P<0.01).The relative expression level of Sirt7.1 gene in ovary decreased with the increase of weekly age,and the relative expression level of Sirt7.1 gene in testis was significantly higher at 2,4 weeks of age than that at 6,8 weeks of age(P<0.05).These results indicated that Sirt1,Sirt2,Sirt6 and their splicosomes had different temporal and spatial expression profiles in mouse gonads,and these different splicosomes may play different functions in different stages of gonad development in male and female mice.
