摘要
目的基于指纹图谱及网络药理学对经典名方甘姜苓术汤进行质量控制及功效关联物质预测分析,为甘姜苓术汤质量控制提供参考依据。方法采用UPLC法建立指纹图谱,运用网络药理学方法构建“成分-靶点-通路”网络,结合化学计量学分析,预测甘姜苓术汤潜在的功效关联物质。结果建立15批甘姜苓术汤指纹图谱,标定16个共有峰,指认8个色谱峰;8个成分主要通过作用于非受体酪氨酸蛋白激酶(SRC)、磷脂酰肌醇3-激酶调节亚基1(PIK3R1)、丝裂原活化蛋白激酶1(MAPK1)等靶点,调控癌症通路、磷脂酰肌醇-3羟激酶-蛋白激酶B(PI3K-Akt)、血管内皮生长因子(VEGF)等信号通路发挥作用;化学计量学方法将样品分为两类,筛选出甘草酸单铵盐、异甘草苷、6-姜烯酚、芹糖甘草苷作为差异性标志物。结论建立的甘姜苓术汤指纹图谱分析方法稳定性好、准确度高,基本体现复方的整体化学信息,筛选出4个成分作为潜在的功效关联物质,为甘姜苓术汤质量的全面控制提供参考。
Objective Based on fingerprint and network pharmacology,the quality control and efficacy related substances prediction of Ganjiang Lingzhu Decoction(甘姜苓术汤)were analyzed,which provides a reference for the quality control of Ganjiang Lingzhu Decoction.Methods UPLC fingerprints of Ganjiang Lingzhu Decoction was established.The“component-target-pathway”network was constructed by network pharmacology,combined with chemometric methods to predict the potential efficacy related substances of Ganjiang Lingzhu Decoction.Results UPLC fingerprints of 15 batches of Ganjiang Lingzhu Decoction were established.A total of 16 common peaks were calibrated.Eight peaks were identified as chromatographic peaks.The 8 components mainly act on SRC,PIK3R1,MAPK1 and other targets to regulate pathways in cancer,PI3K-Akt signaling pathway,VEGF signaling pathway and other signaling pathways.By chemometrics methods,the samples were divided into two categories,and the monoammonium glycyrrhizinate,isoliquiritin,6-shogaol and liquiritin apioside were selected as markers of difference.Conclusion The established fingerprint analysis method of Ganjiang Lingzhu Decoction has strong stability and high accuracy,which basically reflect the overall chemical information of the compound.Four components were selected as potential efficacy related substances to provide reference for the overall quality control of Ganjiang Lingzhu Decoction.
