活血通督汤对大鼠坐骨神经损伤基于Notch信号通路的保护作用

Protective Effects Based on Notch Signal Pathway of Huoxue Tongdu Decoction on Sciatic Nerve Injury in Rats

摘要 目的探究活血通督汤在大鼠坐骨神经损伤中的保护效应。方法选取了54只成年雄性大鼠,按照随机数字表法将其分为假手术组、模型对照组以及活血通督汤组,每组18只。再根据大鼠造模后处死的时间节点,分为术后第7、14、28天三个亚组,每个亚组6只大鼠。假手术组仅进行坐骨神经的暴露而没有进行钳夹处理,而模型组和活血通督汤组则采用钳夹法建立了坐骨神经损伤模型。在术后第7、14、28天分别处死各亚组大鼠并取材。观察大鼠坐骨神经受损程度及NICD和Hes1蛋白的表达水平。结果模型组的坐骨神经组织病理改变明显,随着时间推移未见改善趋势。活血通督汤组的坐骨神经组织病理变化较模型组有明显改善,随着时间推移呈进一步改善趋势。在蛋白表达方面,与模型对照组相比,活血通督汤组在术后各时间节点的NICD和Hes1的表达量都明显增加(P<0.01)。结论活血通督汤可能通过Notch信号转导通路促进坐骨神经损伤的修复。 ObjectiveTo explore the protective effect of Huoxue Tongdu decoction in rat sciatic nerve injury.Methods A total of 54 adult male rats were selected and divided into sham operation group,model control group and Huoxue Tongdu decoction group,with 18 rats in each group.According to the time node of the rats after modeling,they were divided into three subgroups:the 7th,14th and 28th days after surgery,with 6 rats in each subgroup.The sham operation group only performed sciatic nerve exposure without clamping,while the model group and the Huoxue Tongdutang group used the clamping method to establish a sciatic nerve injury model.On the 7th,14th and 28th days after operation,the rats in each subgroup were sacrificed and sampled.The degree of sciatic nerve damage and the expression levels of NICD and Hes1 proteins were observed.Results The histopathological changes of sciatic nerve in the model group were obvious,and there was no improvement trend with the passage of time.The histopathological changes of sciatic nerve in the Huoxue Tongdu decoction group were significantly improved compared with those in the model group,and showed a further improvement trend with the passage of time.In terms of protein expression,compared with the model control group,the expressions of NICD and Hes1 in the Huoxue Tongdu decoction group were significantly increased at each time node after surgery(P<0.01).Conclusions Huoxue Tongdu decoction can help promote the recovery of sciatic nerve injury by the Notch signaling pathway.
