摘要
本研究旨在探求持久性有机挥发物氯苯与α-Fe_(2)O_(3)颗粒在光诱导下的前体污染物与二次产物对A549细胞毒性的影响,以期为两种污染物毒性作用机制及安全性评价提供依据.采用自制反应器在无光、有光条件下生成反应前后两种污染物,从细胞增殖、细胞膜损伤、氧化应激、炎症反应、细胞微观形态等角度探究两者间的毒性效应差异.A549细胞微观形态学显示,污染物通过内吞作用进入细胞质,导致ROS和IL-6水平上升,对细胞增殖、细胞膜损伤呈明显剂量依赖.同时,光诱导后污染物染毒24 h下细胞形态损伤更严重,对A549细胞增殖的抑制作用更强,ROS生成时间提前,100μg·mL^(-1)浓度下ROS和IL-6的生成增多,导致细胞膜损伤情况更严重.本实验为光诱导后污染物毒性增加提供了依据,并强调了需要重新评估大气前体污染物在二次转化阶段所可能产生的毒性影响.
In order to investigate the effect of precursor pollutants and the secondary products on the cytotoxicity of A549 cells by photochemical conversion of persistent organic volatile chlorobenzene withα-Fe_(2)O_(3) particles,with a goal to provide a basis for the mechanism of toxicity and safety evaluation of the two pollutants.The two pollutants were generated in the with/without presence of light in a self-made reactor,and the toxic effects differences between the two were examined from the perspectives of cell proliferation,cell membrane damage,oxidative stress,inflammatory response,and cellular micromorphology.The microscopic morphology of A549 cells showed that the pollutants entered the cytoplasm through endocytosis,which led to the increase of ROS and IL-6 levels,with obvious dose-dependent effects on cell proliferation and cell membrane damage.Meanwhile,the cellular morphology damage was more serious under 24 h of pollutant exposure after photochemical conversion,the inhibition of A549 cell proliferation was stronger,the time of ROS generation was advanced,and the generation of ROS and IL-6 was increased at the concentration of 100μg·mL^(-1),which led to a more serious situation of cell membrane damage.These results provided a basis for the increased toxicity of pollutants after photochemical conversion and highlighted the need to reassess the possible toxic effects of atmospheric precursor pollutants during the secondary transformation phase.
