摘要
目的 利用UPLC-Q-TOF-MS/MS技术解析复方木鸡合剂成分,为复方木鸡合剂质量控制提供依据。方法 复方木鸡合剂采用Agilent Poroshell SB-C_(18)色谱柱(100 mm×4.6 mm,2.7 μm),流动相0.1%甲酸水-乙腈(正);水-乙腈(负),梯度洗脱;流速0.3 mL·min^(-1);柱温30℃;进样量2 μL;采用电喷雾离子源,在正、负离子模式下,通过对照品比对,精确分子质量和二级质谱裂解规律对检测到的化学成分进行鉴定,结合文献报道,确定化学成分结构式。结果 鉴定出复方木鸡合剂中金雀花碱、苦参碱、没食子酸、绿原酸等34个化学成分,其中包括11个生物碱类、11个有机酸类、5个黄酮类、1个萘醌类、1个酚类、1个核苷类、4个其他类化合物,其中以有机酸类和生物碱类为主。结论 本实验阐明复方木鸡合剂中的组成成分,为其药效物质基础、质量控制和作用机制研究提供了实验基础。
Objective To determine the components in compound Muji mixture by UPLC-Q-TOF-MS/MS,and to provide a basis for the quality control of compound Muji mixture.Methods Compound Muji mixture was analyzed with Agilent Poroshell SB-C18 chromatography column(100 mm×4.6 mm,2.7μm)for gradient elution with the mobile phase of 0.1%formic acid water solution-acetonitrile solution-as positive ion modes,water-acetonitrile solution as negative ion modes at a flow rate 0.3 mL·min 1.The column temperature was 30℃and the injection volume was 2μL.Electriospray ion(ESI)source was used in the analysis under the positive and negative ion modes.The detected chemical components were identified via comparison with reference materials,precise molecular weight,and fragmentation pathways by secondary mass spectrometry.Based on literature reports,the structural formula of the chemical components was determined.Results Totally 34 chemical components were identified in compound Muji mixture,including 11 alkaloids,11 organic acids,5 flavonoids,1 naphthoquinone,1 phenolic,1 nucleoside,and 4 other compounds.Organic acids and alkaloids were the main components.Conclusion The compositions in compound Muji mixture are clarified,which provides an experimental basis for its pharmacodynamics,quality control and mechanism of action.
