Hand, foot and mouth disease(HFMD) is a global infectious disease caused by a variety of enteroviruses, which mainly infects children. Recently, the incidence of typical and atypical HFMD caused by coxsackievirus A6(CVA6) has increased significantly in many countries and regions with diverse clinical manifestations. The molecular evolution of CVA6 and the establishment of herd immunity barrier can affect the outbreak and change of pathogen spectrum of HFMD. In this review, the epidemiology, molecular evolution and recombination, amino acid variation, structural analysis and seroepidemiology of CVA6 are reviewed, aiming to provide ideas for the prevention and control of HFMD.
