摘要
设计合成一类可负载淫羊藿苷的多孔碳纳米材料,能够提供一类更高效的淫羊藿苷的靶点“钩钓”固相载体。本文采用软/硬模板法分别合成多孔碳材料,对其进行氨基功能化修饰后负载淫羊藿苷。首先通过氮气等温吸附/脱附测定多孔碳比表面积和孔结构特征,然后借助紫外可见分光光度法考察所合成两种多孔碳的负载性能,之后通过扫描电子显微镜和透射电子显微镜观察多孔碳吸附前后的微观形貌以及利用傅里叶红外光谱考察多孔碳吸附前后的表面化学组成变化,进而判断多孔碳与药物间的相互作用关系;最后借助CCK-8法验证-NH_(2),修饰前后多孔碳的生物相容性,为多孔碳作为靶点“钩钓”载体平台提供前期实验基础。结果表明:-NH,修饰后的多孔碳材料可有效负载淫羊藿苷,并在50μg·mL^(-1)浓度下具有良好的生物相容性。本研究为淫羊藿苷靶点“钩钓”固相载体平台提供了新的研究方向。
In this study,a class of porous carbon nanomaterials capable of loading icarin and serving as a more efficient solid-phase carrier for icarin's"target identification"was designed and synthesized.The porous carbon nanomaterials were synthesized employing both soft and hard template methods,subsequently undergoing amino-functionalization to facilitate icarin loading.Initially,the specific surface area and pore structure characteristics of the porous carbon were determined through nitrogen isothermal adsorption/desorption measurements.Subsequently,the loading performance of the two synthesized porous carbon materials was investigated using ultraviolet-visible spectrophotometry.The micro-morphology of the porous carbon before and after adsorption was observed using scanning electron microscopy(SEM)and transmission electron microscopy(TEM).Fourier transform infrared spectroscopy(FTIR)was employed to analyze changes in the surface chemical composition of the porous carbon before and after adsorption,thus elucidating the interaction between the porous carbon and the drug.Finally,the biocompatibility of the porous carbon before and after-NH_(2) modification was evaluated using the CCK-8 method,providing a preclinical experimental basis for the use of porous carbon as a"target identification"carrier platform.The results demonstrated that the porous carbon material modified with-NH,could effectively load icarin and exhibited good biocompatibility at a concentration of 50μg·mL^(-1).This study offered a novel research direction for the development of icarin's“target identification"”solid-phase carrier platform.
