摘要
孤独症谱系障碍(autism spectrum disorder,ASD)是一种神经发育障碍性疾病,其患病率呈逐年上升趋势,给家庭和社会带来巨大的经济和精神负担。ASD发病机制尚未明确,目前治疗主要以康复行为干预和教育为主。特定神经元环路的功能障碍可能由脑发育相关信号通路功能或结构异常所致。ASD动物模型研究及ASD临床研究均提示炎症信号通路可能在ASD的发病机制中发挥重要作用。目前研究较多的有Notch通路、NF-κB通路、JAK/STAT通路、BDNF-TrkB通路、mTOR通路等信号通路。文章就研究较广的炎症信号通路与ASD发病机制的相关研究进行综述,以期为ASD的靶向治疗提供理论依据。
Autism spectrum disorder(ASD)is a neurodevelopmental disorder with an increasing prevalence year by year,which brings huge economic and mental burden to families and society.The pathogenesis of ASD is not yet clear,and currently the treatment mainly focuses on rehabilitation behavior intervention and education.The dysfunction of specific neuronal circuits may be caused by abnormal function or structure of signal pathways related to brain development.Both animal model study and clinical study of ASD suggest that inflammatory signal pathway may play an important role in the pathogenesis of ASD.At present,there are many literature reports and studies on the Notch pathway,NF-κB pathway,JAK/STAT pathway,BDNF-TrkB pathway,mTOR pathway and others.In this paper,we will review the research on the extensive inflammatory signal pathway and the pathogenesis of ASD,in order to provide theoretical basis for the targeted treatment of ASD.
