摘要
以适配体作为分子识别和结合敏感元件,功能化修饰电极为换能元件,构建无标记型电化学适配体传感器用于血清肌红蛋白定量检测。利用L-半胱氨酸(L-Cys)和羧基化多壁碳纳米管(cMWCNTs)氨基、羧基和巯基分别共价连接5’端修饰氨基适配体和金盘基础电极表面,获得Apt/cMWCNTs/Cys修饰功能化电极,明确其电化学性能。实验结果表明,Mb浓度在0.01~10 ng/mL范围内,电化学传感体系的差分脉冲峰电流值与Mb浓度对数呈现良好的线性关系,相关系数为0.993。cMWCNTs/Cys无标记型电化学适配体传感器具有良好的特异性,可实现血清Mb的定量检测,其检测限LOD为0.016 pg/mL,相对标准偏差RSD为2.09%~3.10%,回收率为94.26%~101.30%。
Aptamers are used as molecular recognition and binding sensitive elements,and the functionalized electrode as the transducer element,a label-free electrochemical aptamer-based sensor was constructed and quantitative detected for serum myoglobin samples.In this study,the amino,carboxyl and sulfhydryl group of L-Cys and carboxylated multi-wall carbon nanotubes(cMWCNTs)were covalently connected to the surface of the 5'modified amino aptamer and the gold disk base electrode,and the Apt/cMWCNTs/Cys modified functional electrode was obtained and electrochemical properties characterization.The results showed that the differential pulse peak current value of the sensing system has a good linear relationship with the logarithm of Mb concentration from 0.01~10 ng/mL,the correlation coefficient was 0.993.The cMWCNTs/Cys label-free electrochemical aptasensor has good specificity for serum myoglobin quantitative detection.The limit of detection(LOD)was 0.016 pg/mL,the relative standard deviation(RSD)was 2.09%~3.10%,and the recovery rate was 94.26%~101.30%.
