摘要
目的探究毛兰素(ER)对卵巢癌细胞恶性进展的调控作用及对CCL2-CCR2信号轴的调控机制。方法培养人卵巢癌细胞SKOV3并分为SKOV3组,ER低浓度(ER-L)组、ER中浓度(ER-M)组、ER高浓度(ER-H)组和ER-H+pcDNA-CCL2组。采用CCK-8法和克隆平板实验检测细胞增殖活性;Transwell实验检测细胞迁移及侵袭能力;流式细胞术检测细胞凋亡;蛋白免疫印迹(Western blot)检测细胞中CCL2-CCR2信号轴相关蛋白的表达。结果与SKOV3组相比,ER-L组、ER-M组和ER-H组细胞存活率、克隆形成数量、迁移及侵袭细胞数量以及细胞中CCL2、CCR2表达水平均降低(P<0.05),细胞凋亡率升高(P<0.05);过表达CCL2削弱了高浓度ER对SKOV3细胞恶性进展的抑制作用(P<0.05)。结论ER能够对卵巢癌细胞的恶性进展起到抑制作用,其作用机制可能与CCL2-CCR2信号轴被抑制有关。
Objective To investigate the regulatory effect of erianin(ER)on the malignant progression of ovarian cancer cells and its regulatory mechanism on the CCL2-CCR2 signaling axis.Methods Human ovarian cancer cells SKOV3were cultured and divided into SKOV3 group,ER low concentration(ER-L)group,ER medium concentration(ER-M)group,ER high concentration(ER-H)group,and ER-H+pcDNA-CCL2 group.CCK-8 method and clone plate assay were used to detect the proliferation activity of cells.Transwell experiment was used to detect the migration and invasion ability of cells;flow cytometry was used to detect the apoptosis rate of cells.Western blot was used to detect the expression of CCL2-CCR2signaling axis related proteins of cells.Results Compared with the SKOV3 group,the cell survival rate,the number of clones formed,the number of migrating and invading cells and the expression levels of CCL2 and CCR2 in ER-L,ER-M and ER-H groups were decreased(P<0.05),and the cell apoptosis rate was increased(P<0.05).Overexpression of CCL2 weakened the inhibitory effect of high concentration ER on malignant progression of SKOV3 cells(P<0.05).Conclusion ER can inhibit the malignant progression of ovarian cancer cells,and its mechanism of action may be related to the inhibition of the CCL2-CCR2signaling axis.
