基于半夏无机元素含量分析的产地区分及健康风险评估

Origin Discrimination and Health Risk Assessment Based on Inorganic Element Content Analysis of Pinelliae Rhizoma

摘要 目的:建立半夏中37种无机元素的电感耦合等离子体-质谱(ICP-MS)分析方法,基于无机元素含量分析不同产地的相关性,并进行健康风险评估。方法:采用ICP-MS法检测半夏中无机元素的含量,结合化学计量法对产地进行区分,计算半夏中5种重金属及有害元素的每日最大耐受摄入量,靶标危害系数和致癌风险,进行健康风险评估。结果:不同产区的半夏所含元素种类差异较小,含量差异较大,甘肃省内和甘肃省外的半夏无机元素含量有显著的地域差异,OPLS-DA和PCA分析方法确定了半夏中Mn、Sr、Tl、Th、Rb、Yb等共17种差异性特征元素,根据这些元素可以将甘肃省内半夏分为一类,甘肃省外半夏分为一类;参考2020版《中国药典》中规定的5种重金属及有害元素的标准,34批半夏中重金属及有害元素均未超标。但半夏的风险评估结果显示34批半夏中As和Pb元素存在一定的健康风险,但不存在因服用半夏原药材而导致的潜在致癌风险。结论:试验建立的分析方法准确且灵敏度较高,适用于半夏中多种无机元素的含量测定,不同产地的半夏中无机元素含量存在差异,差异元素可对不同产地进行区分,所建立的健康风险评估模型可为不同产地半夏中所含无机元素的控制和质量安全提供参考。 Objective:To establish an inductively coupled plasma-mass spectrometry(ICP-MS)method for the analysis of 37 inorganic elements in Pinelliae Rhizoma.Based on the content of inorganic elements,the correlation between different places was analyzed and the health risk was assessed.Methods:The content of inorganic elements in Pinelliae Rhizoma was detected by ICPS-MS,and the origin was differentiated by chemometry.The maximum tolerable daily intakes of five heavy metals and harmful elements in Pinelliae Rhizoma were calculated,and the target hazard coefficient and carcinogenic risk were evaluated for health risk.Result:There was little difference in the types of elements and great difference in the content of Pinelliae Rhizoma in different producing areas.There was a significant regional difference in the content of inorganic elements in Pinelliae Rhizoma between Gansu province and outside Gansu Province.OPLS-DA and PCA analysis methods identified a total of 17 different characteristic elements such as Mn,Sr,Tl,Th,Rb and Yb in Pinelliae Rhizoma.According to these elements,Pinelliae Rhizoma in Gansu Province could be divided into one category and Pinelliae Rhizoma in Gansu Province could be divided into one category.Referring to the standards of five heavy metals and harmful elements stipulated in the 2020 edition of the Chinese Pharmacopoeia,the heavy metals and harmful elements in 34 batches of Pinelliae Rhizoma did not exceed the standard.However,the risk assessment results of Pinelliae Rhizoma showed that there were certain health risks of As and Pb elements in 34 batches of Pinelliae Rhizoma,but there was no potential carcinogenic risk caused by taking the original drug.Conclusion:The analytical method established in this experiment was accurate and sensitive,which was suitable for the determination of various inorganic elements in Pinelliae Rhizoma.The contents of inorganic elements in Pinelliae Rhizoma from different producing areas were different,and the different elements could be distinguished from different producing areas.The established health risk assessment model could provide corresponding reference for the control of heavy metals and food safety in Pinelliae Rhizoma from different producing areas.
