摘要
目的基于生物信息学方法筛选调控Rab23表达以及影响头颈部鳞状细胞癌预后相关的MicroiRNA(miRNA)并验证,为进一步探讨头颈部鳞状细胞癌的发生和侵袭作用机制提供新的研究思路和方法。方法依据miRWalk数据库筛选调控Rab23表达的潜在miRNA,根据TCGA公共数据库和miRWalk数据库筛选影响头颈部鳞状细胞癌生存期的差异表达miRNA,双重筛选交集后确定调控Rab23表达及影响头颈部鳞状细胞癌预后相关的miRNA分子,再通过实时荧光定量聚合酶链反应(RT-qPCR)、蛋白免疫印迹和双荧光素酶实验进行结果验证。结果筛选出250个差异表达miRNA,其中8个是潜在调控Rab23表达的miRNA。hsa-miR-363-3p是既能调控Rab23表达,又可显著影响头颈部鳞状细胞癌预后的miRNA。RT-qPCR、蛋白免疫印迹实验证实hsa-miR-363-3p与Rab23在肿瘤细胞和转染细胞模型中呈负调控;双荧光素酶实验验证hsa-miR-363-3p能够靶向作用于Rab23基因。结论hsa-miR-363-3p参与靶向调控Rab23表达,可能参与了头颈部鳞状细胞癌的侵袭和转移的作用机制。
Objective The screening and validation of MicroRNA(miRNA)related to the regulation of Rab23 and the prognosis of head and neck squamous cell carcinoma(HNSCC)was conducted through bioinformatics method,aiming to explore underlying mechanisms in development and invasion of HNSCC,thereby offering novel research perspectives and methodologies.Methods The screening of the potential miRNAs regulating Rab23 expression were performed using the miRWalk database,and the differentially expressed miRNAs associated with prognosis of HNSCC were obtained from both the TCGA public database and the miRWalk database.The miRNA related to the regulation of Rab23 and the prognosis of HNSCC was identifed by overlapping candidate miRNAs of two groups.The results were subsequently validated using RT-qPCR,Western-Blot,and double luciferase reporter assay.Results A total of 250 differentially expressed miRNAs were identified,including eight miRNAs involved in the regulation of Rab23.The hsa-miR-363-3p was related to the regulation of Rab23 expression,as well as the prognosis of HNSCC.The negative regulation between hsa-miR-363-3p and Rab23 in tumor cells and transfected cell models was confirmed by RT-qPCR and Western-Blot assay.The subsequent double luciferase reporter assay confirmed the binding of hsa-miR-363-3p to Rab23.Conclusion The hsa-miR-363-3p involves in the targeted regulation of Rab23 expression and likely plays a role in the invasion and metastasis of HNSCC.
