乳腺癌患者血清miR-325、miR-641、miR-1307水平与临床病理特征、复发的关系

Relationship between serum miR-325,miR-641,miR-1307 levels and clinicopathological characteristics,recurrence in breast cancer patients

摘要 目的探讨乳腺癌(BC)患者血清微小RNA-325(miR-325)、miR-641、miR-1307表达水平及其与临床病理特征、复发的关系。方法选取西安交通大学第一附属医院2018年3月至2020年3月期间收治的198例BC初诊患者(BC组)作为研究对象;另选取同期于西安交通大学第一附属医院就诊的86例乳腺良性病变患者作为对照组。检测各组血清中miR-325、miR-641、miR-1307的表达情况;Cox回归分析影响BC患者术后复发的风险因素;绘制ROC曲线评估血清miR-325、miR-641、miR-1307表达水平对BC复发的预测价值。结果与对照组相比,BC患者血清miR-325、miR-641表达水平降低,miR-1307表达水平升高(均P<0.05),且三者表达水平与TNM分期、淋巴结转移具有相关性(P<0.05);复发组BC患者血清miR-325、miR-641表达水平低于未复发组,miR-1307表达水平高于未复发组(均P<0.05);miR-1307表达是BC患者术后复发的危险因素,miR-325、miR-641为保护因素(均P<0.05),三者联合预测BC复发的AUC为0.935,联合预测效果优于单独预测(P<0.05)。结论BC患者血清miR-325、miR-641、miR-1307表达水平与TNM分期、淋巴结转移有关,且其表达水平与BC复发密切相关。 Objective To investigate the expression levels of serum microRNA-325(miR-325),miR-641,and miR-1307 in patients with breast cancer(BC)and their relationship with clinicopathological characteristics and recurrence.Methods 198 newly diagnosed BC patients(BC group)admitted to the First Affiliated Hospital of Xi'an Jiaotong University from March 2018 to March 2020 were selected as the research subjects;86 patients with benign breast lesions who were treated at the First Affiliated Hospital of Xi'an Jiaotong University during the same period were selected as the control group.The detect expression of miR-325,miR-641,and miR-1307 in the serum of each group.Cox regression was applied to analyze the risk factors affecting postoperative recurrence in BC patients.ROC curve was plotted to evaluate the predictive value of serum miR-325,miR-641,and miR-1307 expression levels for BC recurrence.Results Compared with the control group,the expression levels of miR-325 and miR-641 in the serum of BC patients reduced,while the expression level of miR-1307 increased(both P<0.05),and the expression levels of the three were correlated with TNM staging and lymph node metastasis(P<0.05).The expression levels of miR-325 and miR-641 in the serum of BC patients in the recurrence group were lower than those in the non recurrence group,and the expression level of miR-1307 was higher than that in the non recurrence group(both P<0.05).While miR-1307 expression was a risk factor for postoperative recurrence in BC patients,miR-325 and miR-641 were protective factors(both P<0.05),the combined prediction of the three for BC recurrence had an AUC of 0.935,which was better than single prediction(P<0.05).Conclusion The expression levels of miR-325,miR-641,and miR-1307 in the serum of BC patients are related to TNM staging and lymph node metastasis,and their expression levels were closely related to BC recurrence.
