摘要
帕金森病是一种中枢神经系统变性疾病,为多基因遗传。因中脑多巴胺代谢失调而引起运动障碍等症状。目前,尽管用于治疗帕金森病的药物是针对该疾病发病机制开发的,但是由于在血脑屏障渗透性方面仍存在着一定的缺陷,导致这些药物的治疗效果不佳。纳米材料为帕金森病的药物治疗提供了解决方案,能够将药物靶向到特定区域,解决药物缺乏特定部位的递送问题。纳米材料具有独特的物理化学特性,可以通过不同的机制穿越血脑屏障。最新研究表明,携带纳米载体和合适配体的治疗药物,有助于改善亲、疏水性药物在大脑中的分布,实现特定部位的药物递送。黑磷是近5年备受关注的新型纳米材料,因其独特结构赋予的性能,包括优越的光热/光动力特性、还原性、高载药能力以及良好的生物相容性等,被广泛应用于生物医药领域研究。在这篇综述中,我们将介绍帕金森病的病理机制以及黑磷纳米片在帕金森病治疗中的适用性。
Parkinson’s disease(PD)is a heritable degenerative disease of the central nervous system.Symptoms such as dyskinesia are caused by dysregulation of mesencephalic dopamine metabolism.Currently,although the drugs used for the treatment of Parkinson’s disease have been developed to target the pathogenesis of the disease,there are still some deficiencies in blood-brain barrier permeability that lead to the poor therapeutic effects of these drugs.And nanomaterials offer a solution to Parkinson’s disease treatment by targeting drugs to specific regions,solving the deficiency in site-specific delivery.Nanomaterials have unique physicochemical properties and can cross the blood-brain barrier by different mechanisms.New research identifies therapeutic drugs carrying nanocarriers and suitable ligands that help improve the distribution of hydrophilic drugs in the brain for site-specific drug delivery.Black phosphorus is a novel nanomaterial that has attracted much attention in the last five years and has been widely used in biomedical research due to its unique structure-endowed properties,including superior photothermal/photodynamic properties,reducing properties,high drug-carrying capacity,and good biocompatibility.In this review,we try to present the mechanism of Parkinson’s disease and the applicability of black phosphorus nanosheets in its treatment.
