摘要
水库对河流生源要素的影响及其生态效应已引起人们的日益关注.本研究探索了全程氨氧化菌在澜沧江深大水库沉积物中的垂直分化现象,选取不同深度沉积物,采用实时荧光定量PCR技术测定了全程氨氧化菌分支A和分支B,以及氨氧化古菌和氨氧化细菌的丰度,并采用高通量测序技术和系统发育分析研究了全程氨氧化菌的群落组成.结果表明,全程氨氧化菌分支B丰度和氨氧化细菌丰度随着沉积物深度增加而显著降低.系统发育分析表明分支B的占比很少,分支A中的Candidatus Nitrospira nitrosa(25.34%)和Nitrospira sp.SG-bin2(14.72%)是全程氨氧化菌中的主要优势物种.但只有Candidatus Nitrospira nitrificans的相对丰度随着深度增加而降低,并受到氨氮的显著影响.Chao1和Richness指数也随着沉积物深度增加而变小,氨氮和pH对α多样性也产生了显著影响.本研究表明全程氨氧化菌分支B在澜沧江水库沉积物中出现了生态位垂直分化,这些分化的环境效应还有待进一步深入研究.
The influence of reservoirs on river biogenic elements and their ecological effects have attracted increasing attention.This study explored the vertical differentiation of complete ammonia-oxidizing bacteria(comammox bacteria)in sediments of the Lancang River deep reservoir.The abundance of comammox bacteria(clade A and clade B),ammonia-oxidizing archaea(AOA),and ammonia-oxidizing bacteria(AOB)was determined by qPCR,and the community composition of comammox bacteria was studied by high-throughput sequencing and phylogenetic analysis.The results indicated that the abundance of comammox clade B and AOB decreased significantly with increasing sediment depth.Interestingly,phylogenetic analysis indicated that clade B only accounted for a very small proportion of the comammox bacteria,while Candidatus Nitrospira nitrosa(25.34%)and Nitrospira sp.SGbin2(14.72%)in clade A were dominant comammox bacterial species.However,only the relative abundance of Candidatus Nitrospira nitrificans decreased with increasing sediment depth and was significantly affected by NH+4-N.Chao1 and Richness indices also declined with increasing sediment depth.NH+4-N and pH exhibited a significant effect onαdiversity.These results suggest that comammox clade B has sediment depth-dependent niche differentiation in the Lancang River,and the environmental effects of the differentiation need to be further studied.
