天然细胞及膜仿生药物递送系统在疾病治疗中的研究进展

摘要 开发天然细胞及膜仿生药物递送系统是利用细胞及其来源的生物膜进行药物递送。本文对基于完整细胞及其衍生的细胞外囊泡和细胞膜的仿生药物递送系统进行了概述。此类递送系统在多种疾病的治疗中得到应用,包括肿瘤、炎症和免疫性疾病、心血管疾病、神经退行性疾病和再生医学等领域。在疾病的治疗中,仿生药物递送系统表现出较高的生物相容性、靶向性,引发更低的宿主免疫反应。然而,较多的研究仍局限于临床前研究,这与此类系统的制备复杂性、体内安全性、临床试验有效性评估等方面的难题相关。本文将对上述系统的作用机制、优缺点、未来发展方向及临床转化进行总结讨论。本文旨在通过对此类仿生药物递送系统的综合分析,为疾病的精准治疗策略提供指导性研究方向。 Developing cell and membrane bioinspired natural delivery systems is to use cells and derived membranes for drug delivery.Bioinspired natural delivery systems based on intact cells,extracellular vesicles,and cell membranes are summarized in this paper.Such delivery systems are applied in the treatment of various diseases,including cancer,inflammation and immune disorders,cardiovascular diseases,neurodegenerative diseases,and regenerative medicine.In disease therapy,bioinspired natural delivery systems exhibit high biocompatibility,targeting specificity,and lower host immune responses.However,most studies are still confined to preclinical research,attributed to challenges such as the complexity of system preparation,in vivo safety,and efficacy evaluation in clinical trials.This paper summarizes the mechanisms,advantages,disadvantages,future directions,and clinical translation of this kind of bionic drug delivery systems,aiming to provide research directions for precision disease treatment strategies.
