摘要
采用包埋法固定化高产果糖基转移酶的黑曲霉全细胞,以海藻酸钠-壳聚糖为载体,分别通过单因素和正交试验优化,获得最佳固定化条件。单因素优化试验结果表明:海藻酸钠最适浓度为2%,菌体包埋量最适为150 g·L^(−1),壳聚糖和CaCl_(2)最适浓度均为4%。正交试验结果表明:海藻酸钠浓度为3%,菌体包埋量为200 g·L^(−1),壳聚糖浓度5%,CaCl_(2)浓度为3%时,酶活回收率可达到最佳的74.7%。验证试验表明该固定化细胞连续进行7个批次合成低聚果糖的催化,仍有96.85%的残余活力,具备很好的应用潜力,为合成低聚果糖提供了新的技术路线。
The whole cells of Aspergillus niger with high yield of fructosyltransferase were immobilized by using the embedding method.Then,the sodium alginate-chitosan was used as the carrier,and the optimum immobilization conditions were obtained by using the single factor and orthogonal experiment optimization,respectively.The singlefactor optimization test results showed that:the optimal concentration of sodium alginate was 2%,the optimal quantity of bacteria embedded was 150 g·L^(−1),and the optimal concentrations of chitosan and CaCl_(2)were both 4%.The results of orthogonal test showed that when the concentration of sodium alginate was 3%,the quantity of bacteria embedded was 200 g·L^(−1),the concentration of chitosan was 5%,and the concentration of CaCl_(2)was 3%,the recovery of enzyme activity could reach the optimal 74.7%.The validation test showed that the immobilized cells were catalyzed for the synthesis of fructooligosaccharides in seven consecutive batches,and still had 96.85%residual activity,which had good potential for application and provided a new technical route for the synthesis of fructooligosaccharides.
