天然低共熔溶剂高效提取金耳多糖的研究

摘要 以金耳子实体为原料,采用低共熔溶剂提取金耳多糖。考察不同组合低共熔溶剂对金耳多糖的提取效果,并与常规的热水浸提法进行比较。经过初步筛选,氯化胆碱和尿素组成的低共熔溶剂体系提取率最高。然后,考察了低共熔溶剂的摩尔比、含水量、液料比、提取时间和提取温度对金耳多糖提取率的影响。通过单因素试验和三因素四水平正交试验确定了金耳多糖的最佳提取条件。结果表明:低共熔溶剂在70℃条件下,以氯化胆碱-尿素的摩尔比为1∶1,低共熔溶剂含水量10%,液料比为70∶1(mL/g),提取时间30 min,金耳多糖得率为33.08%±3.60%,是热水浸提法的7倍。因此,本研究提出了一种简单、绿色、经济、高效的提取金耳多糖的方法,为金耳多糖功能性食品的进一步开发利用提供参考。 Deep eutectic solvents(DESs)were used to extract polysaccharides from the fruiting bodies of Tremella aurantialba.The extraction efficiencies of different combinations of DESs on Tremella aurantialba polysaccharides was investigated and compared with the conventional hot water extraction.After the initial screening,DES system composed of choline chloride and urea showed the highest extraction rate.Thereafter,the effects of molar ratio and water content of DES,material-to-liquid ratio,extraction time,and extraction temperature on the extraction rate of Tremella aurantialba polysaccharides were investigated.The optimum extraction conditions were determined by single-factor experiments and three-factor,four-level orthogonal tests.The results showed that the maximum polysaccharides yield of 33.08%±3.60%was achieved by DES extraction under the condition of 70℃for 30 min,with a choline chloride-urea molar ratio of 1∶1,DES water content of 10%,and a liquid-to-solid ratio of 70∶1(mL/g),which was 7 times higher than that of the hot water extraction.Therefore,this study proposed a simple,environment friendly,economic,and efficient method for extracting Tremella aurantialba polysaccharides,it provides a reference for the further development and utilization of Tremella aurantialba polysaccharide functional foods.
