“腹四针”穴位埋线对单纯性肥胖大鼠糖代谢及AMPK-α、NESFATIN-1表达的影响

Effects of"Abdominal Four Needles"Acupoint Embedding on Glucose Metabolism and Expression of AMPK-αand NESFATIN-1 in Simple Obese Rats

摘要 目的:探讨“腹四针”穴位埋线对单纯性肥胖大鼠糖代谢及AMP激活蛋白激酶-α(AMP-activated protein kinase-α,AMPK-α)、摄食抑制因子1(Nesfatin-1)表达的影响。方法:从80只SD大鼠中随机选取12只作为空白组,剩余大鼠喂养高脂饲料8周建立单纯性肥胖模型。将36只造模成功的大鼠随机分为模型组、针刺组、埋线组,每组12只。针刺组大鼠针刺中脘、关元、双侧天枢穴,埋线组大鼠以相同穴位进行埋线治疗,连续4周。治疗结束后,测量大鼠体质量、体长、空腹血糖(fasting plasma glucose,FPG)以及空腹胰岛素(fasting insulin,FINS)水平。采用Western blot法检测胰腺AMPK-α、Nesfatin-1蛋白表达水平。结果:与空白组比较,模型组大鼠体质量、Lee′s指数升高(P<0.01);与模型组比较,针刺组、埋线组大鼠体质量、Lee′s指数降低(P<0.01);与针刺组比较,埋线组大鼠体质量降低(P<0.01)。与空白组比较,模型组大鼠FPG、FINS、胰岛素抵抗指数升高(P<0.01);与模型组比较,针刺组、埋线组大鼠FPG、FINS、胰岛素抵抗指数降低(P<0.01);与针刺组比较,埋线组大鼠FPG、FINS、胰岛素抵抗指数降低(P<0.05)。与空白组比较,模型组大鼠胰腺湿重增加(P<0.01);与模型组比较,针刺组、埋线组大鼠胰腺湿重减少(P<0.01);与针刺组比较,埋线组大鼠胰腺湿重减少(P<0.05)。与空白组比较,模型组大鼠胰腺AMPK-α、Nesfatin-1蛋白表达水平升高(P<0.01);与模型组比较,针刺组、埋线组大鼠胰腺AMPK-α、Nesfatin-1蛋白表达水平降低(P<0.05)。结论:“腹四针”穴位埋线可改善单纯性肥胖大鼠糖代谢,可能与调节胰腺AMPK-α、Nesfatin-1蛋白表达水平有关。 Objective:To investigate the effects of"abdominal four needles"acupoint embedding on glucose metabolism and the expression of AMP-activated protein kinase-α(AMPK-α)and feeding inhibitory factor 1(Nesfatin-1)in simple obese rats.Methods:12 SD rats were randomly selected as the blank group,and the remaining rats were fed high-fat diet for 8 weeks to establish a simple obesity model.Thirty-six rats who were successfully modeled were randomly divided into model group,acupuncture group,and thread embedding group,with 12 rats in each group.The rats in the acupuncture group were acupunctured in the navel,Guanyuan and Tianshu acupoints,and the rats in the thread embedding group were treated with the same acupuncture points for 4 consecutive weeks.After treatment,body weight,body length,fasting plasma glucose(FPG)and fasting insulin(FINS)levels were measured.Western blot was used to detect the protein expression levels of AMPK-αand Nesfatin-1 in pancreas.Results:Compared with the blank group,the body weight and Lee′s index of the model group increased significantly(P<0.01).Compared with the model group,the body weight and Lee′s index of the acupuncture group and the thread embedding group decreased significantly(P<0.01).Compared with the acupuncture group,the body weight of the rats in the thread embedding group decreased(P<0.01).Compared with the blank group,the FPG,FINS and insulin resistance indexes of the model group increased significantly(P<0.01),compared with the model group,the FPG,FINS and insulin resistance indexes of the acupuncture group and the thread embedding group decreased significantly(P<0.01),and compared with the acupuncture group,the levels of FPG and FINS and insulin resistance index in the thread embedding group decreased significantly(P<0.05).Compared with the blank group,the wet pancreatic weight of rats in the model group increased significantly(P<0.01),compared with the model group,the wet pancreatic weight of rats in the acupuncture group and the thread embedding group decreased significantly(P<0.01),and compared with the acupuncture group,the wet pancreatic weight of rats in the thread embedding group decreased significantly(P<0.05).Compared with the blank group,the protein expression levels of AMPK-αand Nesfatin-1 in the pancreas of the model group increased significantly(P<0.01),and compared with the model group,the protein expression levels of AMPK-αand Nesfatin-1 in the pancreas of the acupuncture group and the thread embedding group decreased significantly(P<0.05).Conclusion:The"abdominal four needles"acupoint embedding can improve glucose metabolism in simple obese rats,which may be related to the regulation of pancreatic AMPK-αand Nesfatin-1 protein expression levels.
