摘要
目的:分析中药复方专利治疗痛风的用药规律,为新方研发提供参考。方法:检录国家专利数据库中治疗痛风的处方数据,运用Excel 2021进行性味归经和频次统计,采用R4.3.1软件进行关联规则、聚类和相关性系数分析,通过Cytoscape 3.8构建中药核心复杂网络,并提取密集中药集群。结果:共纳入专利525项,涉及药物998味,频率位于前4的药物有牛膝、黄柏、威灵仙、土茯苓。性味归经分析显示中药药性以寒性为主,温性其次;药味以苦居首,其次为甘、辛;归经前3位依次为肝、肺、脾经。常用药对有苍术-黄柏、萆薢-土茯苓,角药组合有苍术-牛膝-黄柏、黄柏-薏苡仁-苍术、黄柏-土茯苓-牛膝等。通过复杂聚类分析提取出核心药物组合3组,并挖掘出潜在药物组合2个。结论:中药复方专利治疗痛风以清热利湿为治则,其得出的用药规律及潜在药物组合可为指导临床用药及新药开发奠定基础。
Objective:To analyze the medication rules of Chinese compound medicine patent in the treatment of gout,and to provide reference for the research and development of new prescriptions.Methods:The prescription data for treatment of gout in the National Patent Database were collected,and Excel 2021 was used for statistics of natures,flavors,channel entries and frequency.The association rules,clustering and correlation coefficient were analyzed by R 4.3.1 software.The complex core network of Chinese medicine was constructed by Cytoscape 3.8,and intensive Chinese medicine clusters were extracted.Results:A total of 525 patents were enrolled,involving 998 kinds of Chinese medicine.The top four were Achyranthis Bidentatae Radix,Phellodendri Chinensis Cortex,Clematidis Radix et Rhizoma,and Smilacis Glabrae Rhizoma.The analysis of natures and flavors showed that the Chinese medicines were mainly cold in nature,followed by warm and the Chinese medicines were mainly bitter in flavor,followed by sweet and acrid.The top three channel entries were the liver,lung and spleen.Common medicinal pairs were Atractylodis Rhizoma-Phellodendri Chinensis Cortex,Dioscoreae Hypoglaucae Rhizoma-Smilacis Glabrae Rhizoma;triplet medicine combinations include Atractylodis Rhizoma-Achyranthis Bidentatae Radix-Phellodendri Chinensis Cortex,Phellodendri Chinensis Cortex-Coicis Semen-Atractylodis Rhizoma,Phellodendri Chinensis Cortex-Smilacis Glabrae Rhizoma-Achyranthis Bidentatae Radix.Through complex cluster analysis,three core medicinal combinations were extracted,and two potential medicinal combinations were discovered.Conclusion:The treatment of gout by Chinese compound medicine patent takes clearing heat and draining dampness as its principle.The medication rules and potential medicinal combinations obtained can lay a foundation for guiding clinical medication and developing new medicine.
