摘要
为了解我国南海岛礁土壤中的细菌多样性,本研究利用稀释涂布培养法对南海岛礁土壤中的可培养细菌进行分离和系统进化分析,得到52株菌株,分属于3门4纲4目6科14属,结果表明,南海岛礁土壤中的可培养微生物多样性较为丰富。在门水平上,有厚壁菌门(Firmicutes,84.62%)、变形菌门(Proteobacteria,13.46%)、放线菌门(Actinobacteria,1.92%)3个门;在属水平上,前5属分别为Priestia(32.69%)、Bacillus(21.15%)、Alkalihalobacil⁃lus(7.69%)、Pseudoalteromonas(7.69%)、Fictibacillus(5.77%)。通过16S rDNA遗传多样性序列分析可知,有2株细菌的16S rDNA序列与其模式菌株的最大相似性在97.00%~98.00%之间,说明是潜在的新种资源。
To understand the bacterial diversity in the soil of the South China Sea islands and reefs,the diluted coat‐ing culture method was used to isolate and analyze the phylogeny of culturable bacteria from the soil in the South China Sea.Fifty‐two strains were isolated,which belong to 3 phyla,4 classes,4 orders,6 families and 14 genera.The diversity of culturable microorganisms in the soil of the South China Sea is rich.At the phylum level,there are Firmicutes(84.62%),Proteobacteria(13.46%)and Actinobacteria(1.92%).At the genus level,the top five genera were Priestia(32.69%),Bacillus(21.15%),Alkalihalobacillus(7.69%),Pseudoalteromonas(7.69%)and Fictibacillus(5.77%).Sequence analysis of 16S rDNA ge‐netic diversity showed that two bacterial strains have the maximum similarity between 97.00%and 98.00%of 16S rDNA sequence with their model strains,indicating potential new species resources.
