油莎豆微量营养素含量测定及经口毒理试验的评估

摘要 为明确油莎豆茎豆中微量营养素的种类和含量,评价其经口食用的安全性。本研究以黄淮地区主栽品种豫油莎2号为材料,按照相关国家标准检测茎豆中氨基酸、维生素和矿物质元素的种类和含量;以SPF级SD大鼠为饲喂对象,分别采用10、8、4、2 g/(kg·d)作为14 d急性经口毒性和90 d亚慢性经口毒性试验的处理浓度,通过对大鼠身体各项指标的连续测定,分析油莎豆经口毒性的安全特性。结果表明,油莎豆茎豆中含有16种氨基酸,其中含量最高为精氨酸,其次为谷氨酸、天冬氨酸及赖氨酸,维生素中维生素E和维生素C含量最高,矿物元素中含量最高为镁;根据急性毒性剂量分级标准,油莎豆属实际无毒级。90 d经口毒性试验中各剂量的受试动物的体重、食物利用率、脏器质量、脏器系数均无异常改变;眼部检查未发现异常变化;尿液、血液学、血液生化结果各项指标均在正常范围。各脏器病理组织学检查均未见与受试样品有关的病理改变,且未观察到迟发效应。因此,在本研究条件下,根据GB 15193.13—2015 《食品安全国家标准90天经口毒性试验》判定,油莎豆经口毒性属于安全级别。 In order to clarify the types and contents of nutrient elements in Cyperus esculentus L.stem beans and evaluate the safety of oral test,the main cultivar Yuyousha 2 in Huanghuai area was used as the material to detect the types and contents of amino acids,vitamins and mineral elements in stem beans according to relevant national standards.SD rats of SPF grade were fed with 10,8,4,2 g/(kg·d)as the treatment concentration of 14-day acute oral toxicity and 90-day oral toxicity test.The safety characteristics of C.esculentus L.oral toxicity were analyzed by continuous determination of various indexes of rats.The results indicated that C.esculentus L.stem beans contained 16 kinds of amino acids,of which the highest content was arginine,followed by glutamic acid,aspartic acid and lysine.The contents of vitamin E and vitamin C were the highest in vitamins,and the highest content of mineral elements was magnesium ion.According to the acute toxicity dose grading standard,C.esculentus L.was actually non-toxic.In the 90-day oral toxicity test,there were no abnormal changes in body weight,food utilization rate,organ weight and coefficient of each dose of the tested animals.No abnormal changes were found in eye examination.The indexes of urine,hematology and blood biochemical results were all in the normal range.No pathological changes related to the test samples were found in the histopathological examination of each organ,and no delayed effect was observed.Therefore,under the conditions of this experiment,according to GB 15193.13—2015 national food safety standards,the oral toxicity of C.esculentus L.belongs to the safety level.
