摘要
苦楝(Melia azedarach)是速生、多功能可综合利用的乡土树种。通过分层抽样法、AFLP分子标记以及UPGMA聚类分析从苦楝物候多样性、表型多样性、DNA标记多样性以及子代苗期性状多样性4个方面研究出发,探索苦楝在遗传多样性参数、遗传多样性的梯度变异、遗传多样性分布中心等方面的耦合性,为科学合理利用及保护苦楝种质资源提供理论依据。结果表明苦楝群体内的遗传多样性占79.57%,群体间的遗传多样性占20.43%,在谱带频率总方差的贡献中,群体间占27.26%,群体内占72.74%,苦楝群体绝大多数遗传变异存在于群体内;苦楝表型、AFLP分析中遗传多样性参数呈现以纬向为主、经度为辅的地理变异模式,梯度变异耦合性较好;苦楝表型(亲代)、AFLP分子标记、苗期性状(子代)三者在揭示苦楝遗传分化方面有较好的耦合性;根据以上层次的耦合研究,确定苦楝的遗传多样性中心呈“倾斜带状”,在西北部陇南、渭南一带延续至华中、华东一带。
Melia azedarach is one of the wild autochthonous trees with fast growing and multifunctional and comprehensive utilization.Through stratified sampling,AFLP molecular markers and UPGMA cluster analysis,based on the four aspects of M.azedarach phenological diversity,phenotypic diversity,DNA marker diversity and progeny seedling trait diversity,explore M.azedarach's genetic diversity parameters,genetic diversity gradient variation,genetic diversity distribution center,etc.The coupling of these aspects provides a theoretical basis for scientifically and rationally using and protecting the germplasm resources of M.azedarach.The results showed that the genetic diversity within the M.azedarach population accounted for 79.57%,and the genetic diversity among the populations accounted for 20.43%;and the contributions to the total variance of the band frequency,the contribution rate was 27.26%among populations and 72.74%within populations.Most of the genetic variation of M.azedarach population exists within the population.The phenotype of M.azedarach and the genetic diversity parameters in the AFLP analysis show a geographical variation pattern with latitude as the main and longitude as the supplement,and the gradient variation coupling is better.M.azedarach phenotype(parental generation),AFLP molecular markers,and seedling stage traits(offspring)have a good coupling in revealing the genetic differentiation of M.azedarach.Based on the above-mentioned coupling studies at several levels,it is determined that the genetic diversity center of M.azedarach is in the shape of a"tilted belt",extending from the Longnan and Weinan areas in the northwest to Central and Eastern China.
