摘要
糖尿病是一种复杂的代谢性疾病,其患病率持续升高,对各国医疗保健系统造成重大负担。近年来,随着对线粒体未折叠蛋白反应(UPRmt)机制的深入研究,其治疗糖尿病的作用逐渐成为研究热点。UPRmt是一种在线粒体应激时启动的防御机制,旨在恢复和维持线粒体蛋白质稳态,其在糖尿病的发展中发挥重要作用。UPRmt通过调节线粒体伴侣蛋白和蛋白酶的表达维持线粒体蛋白质稳态,从而改善胰岛素敏感性、胰岛素信号传导和能量代谢,对糖尿病的治疗具有潜在应用价值。本文通过阐述UPRmt的调控机制及其在糖尿病防治中的作用,以期寻找关键分子作为干预目标,为糖尿病治疗提供新策略。
Diabetes mellitus is a complex metabolic disease,its prevalence rate continues to rise,causing a heavy burden on the health care system in various countries.In recent years,with the in-depth study of the mechanism of mitochondrial unfolded protein response(UPRmt),its role in the treatment of diabetes has gradually become a research hotspot.UPRmt is a defense mechanism initiated during mitochondrial stress,which aims to restore and maintain mitochondrial protein homeostasis,and plays an important role in the development of diabetes.UPRmt maintains mitochondrial protein homeostasis by regulating the expression of mitochondrial chaperone protein and protease,thereby improving insulin sensitivity,insulin signal transduction,and energy metabolism,which has potential application value for the treatment of diabetes.This article describes the regulatory mechanism of UPRmt and its role in the prevention and treatment of diabetes,with a view to finding key molecules as the intervention target,and providing new strategies for the treatment of diabetes.
