摘要
目的:板蓝根木脂素中具有丰富的生物活性物质,具有较高的开发价值。本研究对板蓝根木脂素类成分的提取工艺进行优化,提高提取效率,同时对板蓝根总木脂素抗氧化活性进行评价。方法:以浸膏得率、板蓝根总木脂素含量、落叶松树脂醇-4-O-β-D-吡喃葡萄糖苷含量为评价指标,对提取工艺中提取方法、乙醇体积分数、提取液倍量、提取次数和提取时间等条件进行优化,并采用DPPH自由基清除能力测定法、ABTS自由基清除能力测定法和羟自由基清除能力测定法对板蓝根总木脂素抗氧化能力进行评价。结果:板蓝根木脂素的最佳提取工艺为超声提取法,80%乙醇体积分数,10倍量提取液,提取2次,每次提取60 min。体外抗氧化活性实验表明,对DPPH自由基最大清除率为41%,对羟基自由基的最大清除率为55%,对ABTS自由基的最大清除率为64%,达到最大清除率时的质量浓度为0.5 mg/mL。结论:本实验采用的板蓝根木脂素的提取工艺提取效率高,稳定性好,板蓝根总木脂素具有较好的抗氧化活性。
Objective:Lignans from Isatis indigotica are rich in bioactive substances and have high development value.In this study,the extraction process of lignans from Radix isatidis was optimized to improve the extraction efficiency,and the antioxidant activity of total lignans from Radix isatidis was also evaluated.Methods:Taking the extract yield,total lignans content of Radix isatidis and larch resin alcohol-4-O-β-D-glucopyranoside content as evaluation indexes,the conditions of extraction method,ethanol concentration,extract volume,extraction times and extraction time in the extraction process were optimized,and the antioxidant capacity of Radix isatidis lignans was evaluated by DPPH radical scavenging ability measurement method,ABTS radical scavenging ability measurement method and hydroxyl radical scavenging ability measurement method.Results:The optimum extraction process of lignans from Isatis indigotica was ultrasonic extraction,80% ethanol concentration,10 times of extraction solution,twice extraction,and each extraction was 60 min.The antioxidant activity experiment in vitro showed that the maximum scavenging rate of DPPH free radical was 41%.The maximum scavenging rate of hydroxyl radical was 55%,and the maximum scavenging rate of ABTS radical was 64%,and the concentration was 0.5 mg/mL when the maximum scavenging rate was reached.Conclusion:The extraction process of lignans from Radix isatidis adopted in this experiment has high extraction efficiency and good stability,and the total lignans from Radix isatidis have good antioxidant activity.
