昆明裂腹鱼CYP19A基因测序验证与进化分析

摘要 通过对昆明裂腹鱼全长转录组数据解析获得CYP19A基因全长序列,设计引物并进行PCR扩增,直接测序获得准确的CYP19A基因序列,运用生物信息学方法对该序列进行分析。结果表明昆明裂腹鱼CYP19A基因的CDS序列长度为1554 bp,蛋白分子量为58370.70 Da,理论等电点(PI)为8.19,共编码了517个氨基酸;该基因有57个磷酸化位点、无信号肽、无跨膜区;二级结构中ɑ-螺旋、无规则卷曲、β-折叠、β-转角占比分别为47.97%、35.01%、11.61%、5.42%。预测三级结构旋转异构体离群值为1.95%,C-Beta偏差有4个。进化分析表明与鲤鱼亲缘关系相近,与鲢鱼亲缘关系较远。该研究为进一步开展昆明裂腹鱼CYP19A基因的功能研究奠定重要基础。 Through the analysis of the whole transcriptome data of Schizothorax grahami,the full-length sequence of CYP19A gene is obtained.Primers are designed and used for PCR amplification,and the accurate CYP19A gene sequence is directly sequenced.The sequence is then analyzed using bioinformatics methods.The results show that the CDS sequence length of CYP19A gene in Schizothorax grahami is 1554 bp,with a protein molecular weight of 58370.70 Da and a theoretical isoelectric point(PI)of 8.19.It encodes a total of 517 amino acids.This gene contains 57 phosphorylation sites,no signal peptide,and no transmembrane region.In the secondary structure,the proportions ofα-helix,random coil,β-sheet,andβ-turn are 47.97%,35.01%,11.61%,and 5.42%,respectively.The prediction of the tertiary structure reveals a rotamer outlier score of 1.95%and 4 C-Beta deviations.Evolutionary analysis indicates that Schizothorax grahami has a close relationship with common carp and a distant relationship with silver carp.This study lays an important foundation for further research on the function of CYP19A gene in Schizothorax grahami.
