低鱼粉饲料中补充牛磺酸对卵形鲳鲹幼鱼蛋白质和脂质代谢的影响

Effects of Low-fishmeal Diets Supplemented with Taurine on Metabolism of Protein and Lipid in Juvenile Golden Pompano Trachinotus ovatus

摘要 为研究牛磺酸对缓解低鱼粉引起的卵形鲳鲹(Trachinotus ovatus)代谢紊乱的影响,在水温28~32℃和盐度34~36下,将体质量(10.05±0.05)g的卵形鲳鲹幼鱼饲养在深圳大鹏新区海上长1 m×宽1 m×高2 m的网箱中,每个网箱70尾,投喂采用鸡粉、大豆浓缩蛋白、发酵豆粕等动植物蛋白部分替代鱼粉配制的粗蛋白约43%的基础饲料,分别添加0(T0组,对照组)、0.40%(T1组)、0.80%(T2组)、1.20%(T3组)和1.60%(T4组)牛磺酸制成的等氮等脂饲料。56 d的饲养结果显示:(1)与对照组相比,添加0.80%~1.60%牛磺酸显著提高了肌肉、肝脏和血清总蛋白(Total protein,TP)含量,显著降低了血氨(Serum ammonia,SA)和肌肉、肝脏及血清中尿素氮(Urea nitrogen,UN)含量,显著上调肌肉中核糖体蛋白S6激酶(S6K)、磷脂酰肌醇-3-激酶(PI3K)、雷帕霉素靶蛋白(TOR)和4E结合蛋白1(4E-BP1)mRNA的相对表达量,促进了肌肉蛋白质沉积;(2)添加0.80%~1.60%牛磺酸通过显著下调肌肉中过氧化物酶体增殖物激活受体γ(PPARγ)、乙酰辅酶A羧化酶(ACC)和脂肪酸合成酶(FAS)mRNA相对表达量与酶活,抑制脂肪合成代谢,显著上调肌肉中过氧化物酶体增殖物激活受体α(PPARα)、激素敏感性脂肪酶(HSL)和肉毒碱棕榈酰转移酶1(CPT1)的mRNA相对表达量与酶活性,促进肌肉中脂肪酸β氧化反应。综上所述,低鱼粉饲料中添加牛磺酸激活S6K/PI3K/TOR/4E-BP1通路,提高卵形鲳鲹肌肉中蛋白质合成代谢能力;此外牛磺酸可抑制卵形鲳鲹肌肉中脂肪合成代谢,促进脂肪酸β氧化供能,节约蛋白质。 In order to investigate the effect of taurine on alleviating the metabolic disorders of golden pompano Trachinotus ovatus caused by low fishmeal,the golden pompano juveniles with body weight of(10.05±0.05)g was reared in cages with length of 1 m×width×2 m high and height of 2 m in Dapeng New Area,Shenzhen,at stocking density of 70 individuals per cage,and fed basic diet containing about 43%crude protein prepared by replacing fishmeal with animal and plant proteins including chicken meal,soybean protein concentrate and fermented soybean meal and the basic isonitrogenous and isoenergetic diets supplemented with 0(Group T0,control group), 0.40% (Group T1), 0.80% (Group T2), 1.20% (Group T3) and 1.60% (Group T4) of taurine at water temperature of 28-32 ℃ and salinity of 34-36 for 56 d. The results showed that compared with the control group, supplementation with 0.80%-1.60% taurine led to significantly improve the levels of total protein (TP) in muscle, liver, and serum, to significantly reduce the levels of serum ammonia (SA) and urea nitrogen (UN) in muscle, liver, and serum. The relative expression levels of ribosomal protein S6 kinase (S6K), phosphoinositide 3-kinase (PI3K), target of rapamycin (TOR), and eukaryotic initiation factor 4E-binding protein 1 (4E-BP1) mRNA were also significantly up-regulated in muscle, promoting protein deposition in muscle. The lipid anabolism was found to be inhibited in the fish fed the diets supplemented with 0.80%-1.60% taurine by significantly down-regulating the relative mRNA expression levels and enzyme activities of peroxisome proliferator-activated receptors gamma (PPARγ), acetyl-CoA carboxylase (ACC), and fatty acid synthetase (FAS) in muscle, and by significantly up-regulating the relative mRNA expression and enzyme activities of peroxisome proliferator-activated receptors-alpha (PPARα), hormone-sensitive lipase (HSL), and carnitine palmitoyl transferase 1 (CPT1), which promoted β-oxidation of fatty acids in muscle. In summary, taurine supplementation in low fishmeal diets activated the S6K/PI3K/TOR/4E-BP1 pathway and improved protein anabolism in golden pompano muscle;in addition, taurine led to inhibit lipid anabolism and promote fatty acids β-oxidation for energy supply in golden pompano muscle, which in turn conserved protein.
