摘要
在老年群体中,神经退行性疾病如帕金森病(Parkinson’s disease,PD)和阿尔茨海默病(Alzheimer’s disease,AD)的发病率和病死率呈逐年升高趋势。目前,神经退行性疾病的发病机制尚不清楚,但多项研究认为与小胶质细胞和炎症因子密切相关。髓系细胞触发受体(triggering receptor expresses on myeloid cells,TREM)是控制炎症、维持骨稳态、调节神经发育和凝血的细胞表面分子家族。TREM1和TREM2是目前表征最为充分的TREM家族成员,主要在小胶质细胞中表达,并通过调节小胶质细胞的激活及炎症因子的释放,在神经退行性疾病中发挥着重要的作用。本文就TREM在神经退行性疾病中的潜在作用机制进行综述,以期进一步探索神经退行性疾病的有效治疗方法。
IThe incidence and mortality rates of neurodegenerative diseases,such as Parkinson’s disease(PD)and Alzheimer’s disease(AD),are steadily increasing among the elderly population.Although the pathogenesis of neurodegenerative diseases remains unclear,numerous studies suggest a close association with microglia activation and inflammatory cytokines.Triggering receptors expressed on myeloid cells(TREM)are a family of cell surface molecules involved in controlling inflammation,maintaining bone homeostasis,regulating neural development,and coagulation.Among them,TREM1 and TREM2 are the most extensively studied members,predominantly expressed in microglia.They play critical roles in neurodegenerative diseases by modulating microglial activation and the release of inflammatory cytokines.This review explores the potential mechanisms of TREM in neurodegenerative diseases,aiming to provide insights into developing effective treatments for these conditions.
