摘要
目的建立不同产地少花龙葵HPLC指纹图谱,并比较其澳洲茄碱、澳洲茄边碱和澳洲茄胺的含量及不同药用部位(果、茎、叶)之间生物碱的含量差异。方法采用XBridge C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-0.2%磷酸水溶液为流动相进行梯度洗脱,柱温为(25±1)℃,流速为1 mL·min^(-1),检测波长为205 nm,分析6个产地各3批次少花龙葵的HPLC图谱,运用中药色谱指纹图谱相似度软件评价,建立不同产地不同批次少花龙葵特征性成分的HPLC指纹图谱;并对不同产地不同药用部位(果、茎、叶)进行含量测定。结果建立了18批次少花龙葵的指纹图谱,共标定了15个共有峰,并确定2号峰为绿原酸,6号峰为木犀草苷,9号峰为澳洲茄碱,10号峰为澳洲茄边碱,11号峰为澳洲茄胺;建立了少花龙葵中3种生物碱的HPLC含量测定方法,结果6个产地中少花龙葵生物碱含量最高的为越南地区,平均值为4666.39μg·g^(-1);少花龙葵果实、叶、茎中生物碱含量平均值分别为3472.58、1990.67、856.79μg·g^(-1)。结论HPLC指纹图谱可用于少花龙葵质量控制;不同产地不同药用部位少花龙葵生物碱含量有所不同,越南太平省地区的生物碱含量较高;果实中的生物碱成分较丰富,叶次之,茎中的生物碱成分相对较少。
Objective To establish an HPLC fingerprints and determine the content of solasonine,solamargine,and solasodine in Solanum americanum Mill.from different origins;and compare the alkaloid content in different medicinal parts(fruits,stems and leaves).Methods The HPLC of 18 batches of Solanum americanum Mill.from different origins were analyzed on an XBridge C18 column(4.6 mm×250 mm,5μm).Acetonitrile-0.2%aqueous phosphoric acid was used as the mobile phase with a gradient elution at a flow rate of 1 mL·min-1 and a detection wavelength of 205 nm.The HPLC profiles of the 18 batches of Solanum americanum Mill.from 6 origins were analyzed,and the similarity was compared.The content of alkaloids in different medicinal parts(fruits,stems and leaves)of different origins were determined.Results Totally 15 common peaks were calibrated through the fingerprints:peak 2 was identified as chlorogenic acid,peak 6 as lignoceroside,peak 9 as solasonine,peak 10 as solamargine and peak 11 as solasodine.Fingerprints of the 18 batches of Solanum americanum Mill.were established by HPLC.Solanum americanum Mill.from Vietnam region had the highest alkaloid content,with a mean value of 4666.39μg·g^(-1);alkaloid content of the fruits leaves,and stems of Solanum americanum Mill.was 3472.58,1990.67 and 856.79μg·g^(-1),respectively.Conclusion The HPLC fingerprints of Solanum americanum Mill.from different origins help control its quality control;the alkaloid content varies in different parts and origins,with higher alkaloid content in Taiping area,Vietnam;alkaloid content is high in the fruits,followed by the leaves,and the stems.
