苏子-半夏对支气管上皮细胞气道黏液高分泌的影响

摘要 目的:探讨苏子-半夏调控支气管上皮细胞黏液高分泌的机制。方法:制备15%苏子-半夏含药血清、甲泼尼龙溶液,采用10%CSE和20 ng/mL脂多糖溶液构建人支气管上皮细胞黏液高分泌模型。按照药物干预分为正常组、模型组、苏子-半夏组、甲泼尼龙组,电子显微镜下观察细胞形态,采用CCK-8法检测支气管上细胞细胞活性,WB法检测网络药理学预测的表皮生长因子受体(EGFR)、肿瘤坏死因子(TNF-α)的蛋白表达。结果:与正常组比较,模型组细胞活性降低(P﹤0.05),与模型组比较,苏子-半夏组中细胞活性升高(P﹤0.05)。与正常组比较,模型组中EGFR、TNF-α的蛋白表达明显增多(P﹤0.05),提示支气管上皮细胞粘液高分泌模型造模成功。与模型组比较,苏子-半夏组、甲泼尼龙组EGFR、TNF-α蛋白表达明显降低(P﹤0.05)。结论:苏子-半夏可以明显降低EGFR、TNF-α的蛋白表达,其调控支气管上皮细胞黏液高分泌机制可能与EGFR、TNF-α蛋白有关。 Objective:To approach the mechanism of fructus perillae-pinellia ternata regulating airway mucus hypersecretion of bronchial epithelial cells.Methods:15%fructus perillae-pinellia ternata-medicated serum and methylprednisolone solution were prepared,and the model of mucus hypersecretion of human’s bronchial epithelial cells was established by 10%CSE and 20 ng/mL lipopolysaccharide solution.They were divided into normal group,model group,fructus perillae-pinellia ternata group and methylprednisolone group by drug intervention.The cell morphology was observed under electron microscope.The cell activity of bronchial epithelial cells was detected by CCK-8 method.The protein expressions of network pharmacology-predicted epidermal growth factor receptor(EGFR)and tumor necrosis factor(TNF-α)were detected by WB method.Results:The cell activity of model group was decreased compared with normal group(P﹤0.05),while the cell activity of fructus perillae-pinellia ternata group was increased compared with model group(P﹤0.05).The protein expressions of EGFR and TNF-αin model group were significantly increased compared with normal group(P﹤0.05),which prompted that the model of mucus hypersecretion of bronchial epithelial cells was successfully established.Compared with model group,the protein expressions of EGFR and TNF-αin fructus perillae-pinellia ternata group and methylprednisolone group were significantly decreased(P﹤0.05).Conclusion:Fructus perillae-pinellia ternata can significantly reduce the protein expressions of EGFR and TNF-α.Its mechanism of regulating mucus hypersecretion of bronchial epithelial cells may be related to proteins of EGFR and TNF-α.
