摘要
本研究基于“肾脑同治”的中医基础理论,应用网络药理学分析,探讨人参-鹿茸药对防治肾阳虚证的物质基础与分子机制。利用TCMSP平台、Gene Cards数据库获取此药对的活性成分及作用靶点,利用SoFDA平台获取肾阳虚证相关靶点,Metascape数据库获取所涉及的信号通路信息,Cytoscape软件构建“病证-药物-成分-靶点-通路”网络,BioGPS数据库获取靶点在人体各器官中表达情况,联合STRING数据库生物信息学网站进行核心活性成分与关键靶标的筛选并进行分子对接。筛选出人参-鹿茸药对中117种活性成分,其中包括17-β-雌二醇、三磷酸腺苷、十六烷酸等9种核心活性成分,与肾阳虚证相关的150个预测靶点,其中包括APOE、B2M、APP等11个核心预测靶点,在肾、脑、肾上腺中表达情况相似,表达量较高的有APOE、B2M等17个靶点,以及主要与PI3K-Akt信号通路和神经内分泌网络相关。本文应用生信分析研究人参-鹿茸防治肾阳虚症的分子机制,并为其“肾脑同治”理论提供科学依据。
The study based on the theory of"kidney-brain homotherapy",the material basis and molecular mechanism of couplet medicines of ginseng and velvet antler in the prevention and treatment of the syndrome of yang deficiency of kidney were discussed by network pharmacological.TCMSP and GeneCards were used to obtain the active components and targets of the couplet medicines,and SoFDA was used to obtain the related targets of the syndrome of yang deficiency of kidney.Metascape was used to obtain the signaling pathway.Cytoscape was used to construct a"disease-drug-component-target-pathway"network.Obtaining the expression of targets in various organs of the human body through the BioGPS.Combined with String,and other bioinformatics websites to screen core active components and key targets,and then conduct molecular docking.Screened out 117 active ingredients in the couplet medicines of ginseng and velvet antler,including 9 core active ingredients such as 17-beta-estradiol and adenosine triphosphate and hexadecane acid and 150 predicted targets related to the syndrome of yang deficiency of kidney,including 11 core prediction targets such as APOE,B2M,APP.The expression pattern is similar in the kidney,brain,and adrenalgland,with 17 targets such as APOE and B2M being highly expressed.Besides,pathway analysis mainly related to PI3K-Akt signaling pathway and neuroendocrine network.Applying bioinformatics analysis to study the molecular mechanism of ginseng and deer antler in preventing and treating kidney yang deficiency,and providing scientific basis for its theory of"kidney brain simultaneous treatment".
