摘要
为分析作物中的胞外ATP受体蛋白质,通过NCBI数据库获取拟南芥外ATP受体蛋白质L-型凝集素受体样激酶1.9(lecRK-1.9)和L-型凝集素受体样激酶1.5(lecRK-1.5)的氨基酸序列,通过BLAST比对和同源树构建,以油菜和玉米为例,在两种作物中筛选与lecRK-1.9和lecRK-1.5亲缘关系相近的蛋白质;使用ExPASy ProtParam、Cell-PLoc、MEME等工具对这些蛋白质进行理化分析、亚细胞定位和二级结构预测.结果显示,油菜、玉米中的12个候选蛋白质的理论等电点为6.52~8.91,大多为疏水蛋白质;12个蛋白质中有6个蛋白质在质膜上具有定位,12个蛋白质二级结构比例均与拟南芥lecRK-1.9、lecRK-1.5相似,其中有8个蛋白质在保守结构域的位置及顺序上与拟南芥lecRK-1.9、lecRK-1.5具有较高的相似性.这些在油菜、玉米中与拟南芥lecRK-1.9、lecRK-1.5蛋白质在理化性质、亚细胞定位、二级结构以及保守结构域具有高相似性的蛋白质,可作为油菜、玉米中胞外ATP受体蛋白质的参选蛋白质.
To analyze the extracellular ATP receptor proteins in crops,we obtained the amino acid sequences of extracellular ATP receptor proteins of Arabidopsis thaliana,including L-type lectin receptor-like kinase 1.9(lecRK-1.9)and L-type lectin receptor-like kinase 1.5(lecRK-1.5)from the NCBI database.Through the BLAST comparison and the construction of phylogenetic tree,the proteins closely related to the lecRK-1.9 and lecRK-1.5 were screened in rapeseed and maize,both of which were taken as the examples of crops.Physical and chemical characteristics,subcellular localization,and secondary structure prediction of these proteins was performed using the tools like ExPASy ProtParam,CellPLoc and MEME.The results showed that the isoelectric points of these proteins ranged from 6.52 to 8.91,and most of them were hydrophobic proteins.Subcellular localization results indicated that 6 proteins in the 12 candidate in rapeseed and maize were localized on the plasma membrane.The proportion of the secondary structure of these 12 proteins were similar to that of Arabidopsis lecRK-1.9 and lecRK-1.5,and the position and order of the conserved domains of 8 of them showed high similarity to that of the Arabidopsis lecRK-1.9 and lecRK-1.5.These proteins in rapeseed and maize,which exhibit high similarity to Arabidopsis lecRK-1.9 and lecRK-1.5 in terms of physicochemical properties,subcellular localization,secondary structure,and conserved domains,could serve as the candidates of extracellular ATP receptor proteins in rapeseed and maize.
