摘要
目的为黄柏骨伤散的全面质控提供依据,探索黄柏骨伤散的药效成分及作用机制;也为其他中药复方预测Q-Marker提供新的思路。方法采用高效液相色谱(HPLC)法,建立黄柏骨伤散的标准指纹图谱,同时采用网络药理学方法构建黄柏骨伤散中4个关键成分的“成分-靶标-疾病”网络,预测其分子作用机制。结果建立了黄柏骨伤散的指纹图谱,并指认了菊苣酸、小檗碱、黄柏碱和槲皮苷等4个共有峰。网络药理学结果表明黄柏骨伤散中菊苣酸、小檗碱、黄柏碱和槲皮苷等有效成分主要调控TNF、PTGS2、SRC等信号通路发挥跌打损伤、骨折、风湿痹痛、腰肌劳损、腰椎颈椎增生的作用。结论通过指纹图谱结合网络药理学,阐明了黄柏骨伤散治疗跌打损伤、骨折、风湿痹痛、腰肌劳损中10个潜在药效成分和作用机制,为黄柏骨伤散的质量控制提供了潜在的质量标志物以及有效的评价手段。
Objective To provide a comprehensive quality control framework for Huangbai Gushang Powder(HBGSS),to investigate its pharmacologically active components and underlying mechanisms of action,and also to offer new insights into predicting quality markers(Q-Markers)for other traditional Chinese medicine(TCM)compound formulas.Methods High-performance liquid chromatography(HPLC)was employed to establish a standard fingerprint profile for HBGSS Concurrently,network pharmacology was utilized to construct a“component-target-disease”network for four key constituents within the formulation,thereby predicting their molecular mechanisms of action.Results The HPLC analysis successfully established a fingerprint profile for HBGSS,identifying four characteristic peaks corresponding to chicoric acid,berberine,phellodendrine,and quercetin.Network pharmacology revealed that these active components primarily modulate the signaling pathways associated with TNF,PTGS2,and SRC,contributing to the treatment of traumatic injuries,fractures,rheumatism,muscle strain,and osteophyte formation in the lumbar and cervical vertebrae.Conclusion By integrating fingerprint chromatography with network pharmacology,this study elucidates ten potential pharmacologically active compounds and their mechanisms involved in the therapeutic effects of HBGSS on?traumatic injuries,fractures,rheumatism,and muscle strain.These findings offer potential Q-Markers and effective evaluation methods for quality control of HBGSS.
