摘要
目的:探究甲状腺癌(TC)组织微小RNA‑338‑3p(miR‑338‑3p)、T细胞因子4(TCF4)表达水平及与临床病理特征和预后的相关性。方法:选择148例河北北方学院附属第一医院2018年9月—2020年9月收治的TC患者为研究对象,术中切除的癌组织作为TC组,癌旁组织作为癌旁组。采用qRT‑PCR法检测组织中miR‑338‑3p、TCF4表达水平;分析TC组织中miR‑338‑3p、TCF4表达水平与临床病理特征及TC患者预后的关系;用多因素Cox回归分析影响TC患者预后的因素。结果:TC组miR‑338‑3p表达水平均明显低于癌旁组,TCF4表达水平均明显高于癌旁组(P<0.05)。Ⅲ~Ⅳ期、低分化和发生淋巴结转移的TC患者miR‑338‑3p低表达、TCF4高表达比例明显高于Ⅰ~Ⅱ期、中高分化和未发生淋巴结转移的TC患者(P<0.05)。TC组织miR‑338‑3p低表达患者3年生存率(45.95%)低于miR‑338‑3p高表达患者(74.32%)(χ2=18.781,P<0.001),TC组织TCF4高表达患者3年生存率(41.89%)低于TCF4低表达患者(78.38%)(χ2=27.978,P<0.001)。miR‑338‑3p、TCF4、临床分期、淋巴结转移是影响TC患者预后的相关因素(P<0.05)。结论:TC组织miR‑338‑3p表达水平明显降低,TCF4表达水平明显升高,其与TC患者临床分期、淋巴结转移有着紧密联系,是影响TC患者预后的相关因素。
Objective:To investigate the correlation between the expression levels of microRNA‑338‑3p(miR‑338‑3p)and T cell factor 4(TCF4)in thyroid cancer(TC)tissue with clinical pathological features and prognosis.Methods:From September 2018 to September 2020,148 TC patients admitted to The First Affiliated Hospital of Hebei North University were involved in this study,their intraoperative resections of TC samples and adjacent tissues were acquired.qRT‑PCR method was applied to detect the expression levels of miR‑338‑3p and TCF4 in tissues.The relationship between the expression levels of miR‑338‑3p and TCF4 in TC tissue and clinical pathological features and prognosis was analyzed.The factors affecting the prognosis of TC patients were analyzed using multivariate Cox regression.Results:The expression levels of miR‑338‑3p in the TC samples were lower than those in the adjacent tissues,and the expression level of TCF4 was higher(both P<0.05).The proportions of miR‑338‑3p low expression and TCF4 high expression in TC patients with stageⅢ‑Ⅳ,low differentiation,and lymph node metastasis,respectively,were higher than in stageⅠ‑Ⅱ,medium to high differentiation,and no lymph node metastasis,respectively(all P<0.05).The 3‑year survival rate(45.95%)of patients with low expression of miR‑338‑3p in TC tissue was lower than that of patients with high expression of miR‑338‑3p(74.32%)(χ2=18.781,P<0.001),and the 3‑year survival rate(41.89%)of patients with high expression of TCF4 in TC tissue was lower than that of patients with low expression of TCF4(78.38%)(χ2=27.978,P<0.001).miR‑338‑3p,TCF4,clinical stage,and lymph node metastasis were related factors that affected the prognosis of TC patients(P<0.05).Conclusion:The expression level of miR‑338‑3p in TC tissue is obviously reduced,while the expression level of TCF4 is obviously increased.They are closely related to the clinical stage and lymph node metastasis in TC patients,and are related factors affecting the prognosis of TC patients.
