赶黄草茎UPLC指纹图谱建立及其成分分析

摘要 基于超高效液相色谱法(UPLC)指纹图谱,结合成分含量并采用化学模式识别法、聚类热图分析及正交偏最小二乘法-判别分析(OPLS-DA)综合评价不同产地赶黄草茎的质量。结果表明:建立的赶黄草茎UPLC指纹图谱共标定6个共有峰,分别是紫云英苷、乔松素-7-O-葡萄糖苷、槲皮素、山奈酚、乔松素-7-O-(3″-O-没食子酰基-4″,6″-六羟基联苯二甲酰基)-β-葡萄糖苷(PGHG)和赶黄草茎苷A;16批赶黄草茎相似度为0.777~0.998;16批赶黄草茎样品中14种化合物含量之间有显著差异。通过聚类热图分析将16批赶黄草茎样品分为3类,四川省泸州市古蔺县样品与其他产地样品能被完全区分;OPLS-DA共筛选出9个质量差异标志性成分。建立的赶黄草茎UPLC指纹图谱及14个成分含量测定方法可靠、重复性好,其化学模式识别方法稳定可行,可为赶黄草茎质量研究提供参考。 Based on the fingerprint of ultra-high performance liquid chromatography(UPLC),combined with the content of components,the quality of Penthorum chinense Pursh stem from different producing areas was comprehensively evaluated by chemical pattern recognition,cluster heat map analysis and orthogonal partial least squares-discriminant analysis(OPLS-DA).The results showed that there were six common peaks in the UPLC fingerprint of Penthorum chinense Pursh stem,which were astragalin,pinocembrin-7-O-glucoside,quercetin,kaempferol,PGHG and thonningianin A.The similarity of 16 batches of Penthorum chinense Pursh stem was 0.777-0.998.There were significant differences in the contents of 14 compounds among 16 batches of Penthorum chinense stem samples.The 16 batches of Penthorum chinense Pursh stem samples were divided into three categories by cluster heat map analysis.The samples from Gulin County,Luzhou City,Sichuan Province could be completely distinguished from the samples from other producing areas.A total of 9 quality difference marker components were screened out by OPLSDA.The established UPLC fingerprint of Penthorum chinense Pursh stem and content determination method of 14 components were reliable and reproducible.The chemical pattern recognition method was stable and feasible,which could provide reference for the quality study of Penthorum chinense Pursh stem.
