黄芪甲苷对淮南麻黄鸡生长性能及血清生化指标、抗氧化能力的影响

Effect of Astragaloside IV on growth efficiency,serum biochemical index,and hepatic antioxidant potential in Huainan partridge chicken

摘要 本文旨在探究黄芪甲苷(Astragaloside IV,ASIV)对淮南麻黄鸡生长性能、血清生化及肝脏抗氧化性能的影响,及其抗氧化作用机制。选取180只1日龄健康淮南麻黄鸡,随机分为3组:对照组(CON)、50 mg/kg ASIV(L-ASIV)和100 mg/kg ASIV(H-ASIV)组,每组6个重复,每个重复10只。于试验14、21、42 d分别测定血清生化和肝脏抗氧化指标。网络药理学获取ASIV抗氧化作用靶点,并对相关靶点进行GO与KEGG富集分析。与CON组相比,L-ASIV和H-ASIV试验组终末体质量和平均日增重升高。试验14 d时,L-ASIV组谷胱甘肽(Glutathione,GSH)和总抗氧化能力(Total Antioxidant Capacity,T-AOC)显著升高(P<0.05),血清碱性磷酸酶(Alkaline Phosphatase,ALP)和总胆红素(Total Bilirubin,TB)以及间接胆红素(Indirect Bilirubin,IB)显著降低(P<0.05)。H-ASIV组过氧化氢酶(Catalase,CAT)显著升高,而血清ALP、天门冬氨酸氨基转氨酶(Aspartate Aminotransferase,AST)、直接胆红素(Direct Bilirubin,DB)、TB、IB以及乳酸脱氢酶(Lactate Dehydrogenase,LDH)显著降低(P<0.05);21 d时,L-ASIV组GSH显著升高(P<0.05),H-ASIV组T-SOD显著升高(P<0.05),TB和IB显著降低(P<0.05);42 d时,在L-ASIV组中,T-SOD、GSH、T-AOC及CAT酶活性均显著上升(P<0.05),同时MDA含量显著降低(P<0.05),H-ASIV组,T-AOC、GSH与CAT酶活性亦显著增强(P<0.05),MDA含量显著降低(P<0.05)。网络药理学预测ASIV抗氧化潜在作用靶点40个,这些靶点可富集到19个信号通路。研究证明ASIV能显著提高淮南麻黄鸡的体质量,对于血清生化以及抗氧化能力产生一定的有益影响,在本试验条件下建议ASIV的添加量为50 mg/kg·BW,ASIV抗氧化作用可能与其作用于STAT3、IL6、CASP3、BCL2、IL1β等靶点影响丝裂原活化蛋白激酶(Mitogen Activated Protein Kinases,MAPK)、细胞凋亡等相关。 The aim of this study was to examin the impact of Astragaloside IV(ASIV)on growth performance,serum biochemical parameters,and antioxidant properties of liver tissues in Huainan partridge chicken,and the mechanism underlying its antioxidant action,which provides a reference for the application of ASIV in poultry farming.One hundred and eighty healthy 1-day-old Huainan partridge chickens were randomly assigned to three groups:control(CON),50 mg/kg ASIV(L-ASIV),and 100 mg/kg ASIV(H-ASIV),with six replicates per group and ten birds per replicate.Growth performance,serum biochemistry and antioxidant indices were determined at 14 d,21 d and 42 d of the experiment.The targets of antioxidants of ASIV were obtained through network pharmacology,and the related targets were enriched by GO and KEGG.In the experimental group,the final body weight and average daily weight gain were significantly greater than those observed in the CON group.On the 14th day of the experiment,hepatic glutathione(GSH)and total antioxidant capacity(T-AOC)were significant increased(P<0.05).In contrast,serum levels of alkaline phosphatase(ALP),total bilirubin(TB),and indirect bilirubin(IB)were significantly reduced(P<0.05)in the L-ASIV group.The levels of catalase(CAT)were significantly elevated in the H-ASIV group,while serum ALP,AST,direct bilirubin(DBIL),TB,IB,and lactate dehydrogenase(LDH)exhibited significant decrease(P<0.05).On day 21,GSH levels were significantly elevated in the L-ASIV group(P<0.05),while T-SOD activity showed a significant increase in the H-ASIV group(P<0.05).TB and IB levels were significantly reduced(P<0.05).On day 42,the levels of T-SOD,GSH,T-AOC,and CAT were significantly elevated(P<0.05)in the L-ASIV group,while MDA activity was markedly diminished(P<0.05).In the H-ASIV group,T-AOC,GSH and CAT levels were significantly higher(P<0.05)and MDA activity was significantly lower(P<0.05).Network pharmacology predicted 40 potential antioxidant targets of ASIV,and these targets could be enriched to 19 signaling pathways.ASIV significantly improves the body weight of Huainan partridge chickens,and has beneficial effects on serum biochemistry and antioxidant capacity.The suggested dosage of ASIV is 50 mg/kg body weight.The anti-oxidation effect of ASIV may be related to the effect of ASIV on MAPK,apoptosis and other targets such as STAT3,IL6,CASP3,BCL2 and IL1β.
