核因子κB抑制剂parthenolide联合柔红霉素对髓系白血病小鼠的治疗效应

Efficacy of NF-κB inhibitor parthenolide in combination with daunorubicin in mice with myeloid leukemia

摘要 目的:探讨核因子κB(NF-κB)抑制剂parthenolide(PN)联合柔红霉素(DNR)对髓系白血病小鼠的治疗效应。方法:20只Nu/Nu裸鼠通过接种急性单核细胞白血病细胞株THP-1构建移植瘤模型后,采用随机数字表法分为空白对照组、PN组、DNR组和PN+DNR组,每组5只。PN组小鼠每2 d腹腔注射0.2μg PN,共7次;DNR组小鼠分别于第1、2、3、8、9、10天腹腔注射0.6 mg DNR;PN+DNR组给药方案同各单药组;空白对照组只注射等体积PBS。第16天处死小鼠,分离瘤体,称取质量,计算瘤体积及抑瘤率;采用TUNEL法检测移植瘤组织细胞凋亡率。结果:处理第16天,空白对照组、PN组、DNR组和PN+DNR组的瘤体积变化值分别为(1365.13±426.79)、(695.68±129.29)、(117.68±58.36)、(-44.55±78.74)mm~3,PN和DNR均可抑制瘤体积(P<0.05),二者联合有协同作用(P=0.034);PN、DNR组、PN+DNR组的抑瘤率分别为32.5%、71.9%、86.8%。4组瘤细胞凋亡率分别为(4.50±1.58)%、(13.20±3.36)%、(34.32±3.40)%和(36.62±3.56)%,PN和DNR均可诱导瘤细胞凋亡,二者有协同作用(P=0.038)。结论:PN有抗髓系白血病效应,PN和DNR联合有协同作用。 Aim:To explore anti-leukemic efficacy of nuclear factor kappa B(NF-κB)inhibitor parthenolide(PN)in combination with daunorubicin(DNR)in mice with myeloid leukemia.Methods:Twenty nude mice were used to construct xenograft model by inoculating acute monocytic leukemia THP-1 cells and randomly assigned into blank control group(treated with PBS),PN group,DNR group,and PN+DNR group,5 mice for per group.The mice in PN group were treated with 0.2μg PN per mouse,intraperitoneal injected once every 2 days within 14 days.The mice in DNR group were treated with 0.6 mg DNR,injected on day 1,2,3,8,9 and 10.The mice in PN+DNR group were treated with combination of PN and DNR,using the same treatment regimen as above.All mice were executed on day 16 and tumors were separated and weighted,and the anti-tumor rate in each group was calculated.Cell apoptosis in the separated tumor tissue was tested using TUNEL assay.Results:On day 16,the changes of tumor volumes of blank control group,PN group,DNR group,and PN+DNR group were(1365.13±426.79),(695.68±129.29),(117.68±58.36),and(-44.55±78.74)mm~3,respectively;both PN and DNR could inhibit tumor volume,and there was a synergistic effect when they were combined(P=0.034).The tumor inhibition rates of PN group,DNR group,and PN+DNR group were 32.5%,71.9%,and 86.8%,respectively.The cell apoptosis rates of the above groups were(4.50±1.58)%,(13.20±3.36)%,(34.32±3.40)%,and(36.62±3.56)%,respectively;both PN and DNR could induce the apoptosis of tumor cells,and there was a synergistic effect between them(P=0.038).Conclusion:PN has anti-myeloid leukemia effects,and PN and DNR have a synergistic effect in combination.
