摘要
产肠毒素性大肠埃希菌(ETEC)是导致人畜腹泻的主要病原菌.本文以苦杏仁粕为原料,将其1%的提取液与ETEC H10407共同培养,研究苦杏仁粕对ETEC生长、形态等的影响,并利用转录组测序技术,探究苦杏仁粕提取液对ETEC中基因的转录表达的作用.结果表明:1%的苦杏仁粕提取液可以显著抑制ETEC的生长,破坏菌体细胞形态,减少了脂肪酸合成、蛋白质翻译通路相关基因的表达,增加了细菌趋化性信号通路相关基因cheB、cheR,ABC转运蛋白途径中铁元素转运相关基因fecB、fecC、fecD等的表达.苦杏仁粕可能引发了ETEC的代谢紊乱,从而抑制了其生长.研究结果将为开发利用苦杏仁,用于ETEC的防治提供参考.
In human and farm animals,Enterohaemorrhagic Escherichia coli(ETEC)is the main pathogen causing diarrhea.In this study,bitter almond meal at 1%was co-cultivated with ETEC,to study the effect of bitter almond meal on ETEC in term of bacterial growth and morphology.Transcriptome sequencing technology was also used to explore the effect of bitter almond meal on the transcriptional expressions of genes in ETEC.This research found that 1%bitter almond meal could significantly inhibit the growth of ETEC,destroy bacterial cell morphology,inhibit the expressions of genes related to fatty acid synthesis and protein translation pathways,and promote expressions of genes related to bacterial chemotaxis signaling pathway such as cheB and cheR,and iron ion transport system in the ABC transport protein pathway such as fecB、fecC and fecD.Bitter almond meal probably caused metabolic disorders in ETEC,thereby inhibiting its growth.Results of this study will provide reference for the development and utilization of bitter almond meal to prevent and treat ETEC.
