摘要
目的探讨长链非编码RNA叉头盒转录基因D2反义RNA1(LncRNA FOXD2-AS1)通过调节miR-338-3p/含蛋白1的Ankyrin重复序列和BTB/POZ结构域(ABTB1)轴对结直肠癌细胞增殖、迁移和侵袭的影响。方法应用实时荧光定量PCR(qRT-PCR)检测结直肠癌患者组织以及细胞系(SW620、HCT116、SW480)中LncRNA FOXD2-AS1和miR-338-3p的表达水平,Western blot检测ABTB1、增殖细胞核抗原(PCNA)、基质金属蛋白酶(MMP)-9、MMP-2等蛋白的表达,通过克隆形成实验和CCK-8实验进行评估细胞增殖能力,通过划痕实验和Transwell实验检测细胞迁移和侵袭能力,通过miR-338-3p抑制实验进一步分析miR-338-3p对LncRNA FOXD2-AS1沉默后细胞增殖、迁移和侵袭的逆转作用,同时验证LncRNA FOXD2-AS1与miR-338-3p、miR-338-3p与ABTB1的相互关系。结果在结直肠癌组织和细胞系中,LncRNA FOXD2-AS1和ABTB1蛋白表达上调,miR-338-3p表达下调;SW480细胞中的LncRNA FOXD2-AS1和ABTB1表达最高,miR-338-3p表达最低。沉默LncRNA FOXD2-AS1的SW480细胞其细胞增殖、迁移和侵袭显著下降,miR-338-3p表达升高,ABTB1、MMP-9、PCNA、MMP-2蛋白水平下调。miR-338-3p抑制实验表明,miR-338-3p的抑制可逆转LncRNA FOXD2-AS1沉默对细胞增殖、迁移和侵袭的抑制作用及对ABTB1、MMP-9、PCNA、MMP-2蛋白水平的下调作用。结论沉默LncRNA FOXD2-AS1可通过调节miR-338-3p/ABTB1轴抑制结直肠癌细胞增殖、迁移和侵袭。
Objective To investigate the effect of long non-coding RNA forkhead box transcription factor D2 antisense RNA 1(LncRNA FOXD2-AS1)on the proliferation,migration,and invasion of colorectal cancer cells by regulating the miR-338-3p/ankyrin repeat domain and BTB/POZ domain containing protein 1(ABTB1)axis.Methods The expression levels of LncRNA FOXD2-AS1 and miR-338-3p in colorectal cancer patient tissues and cell lines(SW620,HCT116,SW480)were measured by quantitative real-time PCR(qRT-PCR).The protein levels of ABTB1,proliferating cell nuclear antigen(PCNA),matrix metalloproteinase(MMP)-9,and MMP-2 were detected by Western blot.The proliferation ability of cells was evaluated by colony formation and CCK-8 assays.The migration and invasion abilities of cells were assessed by scratch wound healing and Transwell assays.Furthermore,the role of miR-338-3p in reversing the effects of LncRNA FOXD2-AS1 silencing on cell proliferation,migration,and invasion was analyzed by miR-338-3p inhibition experiments.The interactions between LncRNA FOXD2-AS1 and miR-338-3p,and between miR-338-3p and ABTB1,were also validated.Results In colorectal cancer tissues and cell lines,the expression of LncRNA FOXD2-AS1 and ABTB1 protein was upregulated,while miR-338-3p expression was downregulated.The highest expression of LncRNA FOXD2-AS1 and ABTB1,and the lowest expression of miR-338-3p,was observed in SW480 cells.Silencing LncRNA FOXD2-AS1 in SW480 cells significantly reduced cell proliferation,migration,and invasion,increased miR-338-3p expression,and downregulated ABTB1,MMP-9,PCNA,and MMP-2 protein levels.miR-338-3p inhibition experiments showed that the suppression of miR-338-3p could reverse the inhibitory effects of LncRNA FOXD2-AS1 silencing on cell proliferation,migration,and invasion,as well as the downregulation of ABTB1,MMP-9,PCNA,and MMP-2 protein levels.Conclusion LncRNA FOXD2-AS1 may promote the proliferation,migration,and invasion of colorectal cancer cells by regulating the miR-338-3p/ABTB1 axis.
