摘要
目的 研究哮喘与非哮喘中肥大细胞脱颗粒。方法 2 9名轻度哮喘患者参加了本项研究。支气管肺泡灌洗液 (BALF)中的细胞经冲洗后 ,加入抗IgE抗体或CI或缓冲液的试管中进行激发试验。组织中肥大细胞的悬浮过程由Ⅰ型胶原酶和Ⅰ型透明质酸酶在 37℃水浴箱中完成。BALF中的组胺采用酶联免疫反应试剂盒测定 ,而组织中的组胺则采用以玻璃纤维为基础的荧光方法来测定。结果 哮喘患者BALF中的肥大细胞对抗IgE抗体的敏感性要比非哮喘者至少敏感 10 0倍 ;对CI刺激的敏感性与非哮喘者的相似。
Objectives To investigate mast cell degranulation in as th matics and non asthmatics.Methods29 mild asthmatics were sele cted for the study. After being washing,the cells from bronchoalveolar lavage f luid (BALF) were added to tubes containing anti IgE or CI for experiments of ac tivation. Cells from tissues were dispersed with collagenase and hyaluronidase a nd finished in 37 ℃ water bath. An enzyme linked immunoassay was employed to detect histamine released from BALF mast cells. A glass fibre based, histamin e fluorometric assay was used to measure histamine in supernatants of dispersed mast cells.Results The mast cells from BALF of asthmatics were at least 100 fold more sensitive to anti IgE than the mast cells from non asth matics. Both asthmatics and non asthmatics had similar response to CI stimulati on. Conclusion Mast cells in asthmatic BALF had enhanced sensit ivity to IgE dependent stimulation.
