黄芪毛状根多糖与黄芪多糖化学组成及免疫活性的比较 被引量:42

摘要 目的 比较生物技术培养的黄芪毛状根中多糖与栽培黄芪中多糖的化学组成和免疫活性。方法 采用乙醇分级沉淀和凝胶渗透柱层析分离纯化多糖 ;硫酸 -苯酚法测定多糖含量 ;完全酸水解、乙酰化、气相色谱测定单糖组成 ;采用免疫器官重量 ,腹腔巨噬细胞吞噬功能 ,溶血素生成 ,〔3H〕- Td R掺入 ,诱生 IFN- γ的含量测定等方法研究免疫活性。结果 黄芪毛状根中多糖为 1.85 % ,黄芪中多糖为 2 .6 1%。黄芪毛状根多糖 (HAPS)由鼠李糖、阿拉伯糖、木糖、甘露糖、半乳糖和葡萄糖组成 ,摩尔比为 1∶ 2 .2 6∶ 0 .2 1∶ 0 .74∶ 2 .4 9∶ 19.4 7。黄芪多糖 (APS)组成与 HAPS相同 ,摩尔比为 1∶ 4 .34∶ 3.92∶ 1.95∶ 11.4 1∶ 2 0 .5 2。采用乙醇分级沉淀各自获得 3个部分多糖 ,经DEAE- Sepharose FF柱层析表明 :HAPSI和 APSI,HAPSII和 APSII图谱相似 ,HAPSIII与 APSIII有一定的差异。 HAPS和 APS均能显著增加小鼠脾脏重量 ,HAPS能提高小鼠胸腺重量 ;二者均增加小鼠腹腔巨噬细胞数量 ,APS显著促进小鼠腹腔巨噬细胞巨噬功能 ;HAPS和 APS均抑制小鼠溶血素生成 ;促进小鼠脾淋巴细胞增殖 ;APS使脾淋巴细胞诱生 IFN- γ的作用高于 HAPS。结论  HAPS和 APS化学组成相似 ,二者具有相同的免疫活性 ,只是强度略有差异。 Object To compare the compositions and the immunoactivities of polysaccharide in hair root of Astragalus membranaceus (Fisch.) Bge.(HAPS), which was cultivated by biotechnology, with the polysaccharide in A. membranaceus (APS), to prove the similar properties both of them. Methods Sulphuric acid-phenol method was used to analyze the polysaccharide contents; completely acid hydrolysis, aditol acetate derivative and GC were used to determine the polysaccharides; the weight of spleen and thymus; phagocytosis function of macrophage ; hemolysin production and 〔 3 H〕-TdR incorperation ELISA kit assays were used to evaluate the immunoactivities in mice. Results The polysaccharides of the hair root were 1.85%, that of A. membranaceus were 2.61%. Both of HAPS and APS had the same monosaccharide composition, but the molar ratio of them was different. HAPS and APS were divided into three parts by different ethanol concentration. The homogeneity was verified by DEAE-Sepharose FF chromatography. HAPS I and APS I were similar, HAPS II and APS II were also similar, HAPS III and APS III were different. HAPS and APS increased the weight of mouse spleen, APS increased the weight of thymus. Both of them increased phagcytosis function of macrophage and inhibited the hemolysin production in mice. HAPS and APS significantly stimulated lymphocyte proliferation of spleen and secreted IFN-γ. ConclusionThe results confirmed that HAPS and APS had similar chemical compositions and immunoactivities, but the intensity showed slightly difference
