摘要
以 2 1份玉米骨干自交系及其组配的 13个杂交种为材料 ,进行 SSR标记分析。从 2 2 0对引物中 ,筛选出扩增带型稳定、多态性丰富的 5 8对引物 ;杂交种的 SSR图谱基本上表现为双亲互补带型。利用两个或三个引物组合构建的SSR图谱 ,通过统计测验可以将 13个杂交种区分。实验表明 ,应用 SSR标记技术结合单籽粒 DNA快速提取方法 ,可以快捷。
The polymorphism of 21 maize elite inbred lines and 13 hybrids were analyzed with SSR marker system. Fifty eight out of 220 pairs of SSR primers were selected, which gave stable and polymorphic amplification profiles. Basically, the amplification bands of hybrids were complementary with that of the parents. Thirteen hybrids could be distinguished by combinations of two or three primers selected and statistical analysis. The rapid DNA extracting procedure from single kernel was established. The experiment showed that SSR technique could be effectively exploited in hybrid seed purity test of maize.
