10-羧基喜树碱对人膀胱癌细胞染色质DNA的断裂作用的研究

摘要 目的 :探讨 10 -羧基喜树碱 (10 -Hydroxylcamptothecine,10 -HCPT)对T2 4人膀胱癌细胞染色质DNA的断裂作用。方法 :采用快速、灵敏的检测细胞DNA损伤的单细胞凝胶电泳 (Singecellgelelectrophoresis,SCGE)技术 ,观察不同浓度 10 -HCPT (0 0 1~ 0 1mg/ml)分别作用于T2 4人膀胱癌细胞不同时间 (2 4h和 4 8h)后细胞DNA的损伤情况。以“彗星样”淋巴细胞出现率 (计数一定量细胞其中彗星样细胞所占的比例 )、总彗星长度(“彗星”电泳方向上的最大长度 ,即尾长 )和尾距 (尾部DNA占总DNA的百分比与头尾部中心间距的乘积 )来评价淋巴细胞DNA的损伤程度。结果 :10 -HCPT能剂量依赖性地导致T2 4人膀胱癌细胞DNA的明显断裂损伤 ,能使“彗星样”膀胱癌细胞出现率、总彗星长度和尾距显著增加 (P <0 0 5 ,P <0 0 1) ,并随着时间的延长损伤加重。结论 :HCPT对膀胱癌细胞具有很强的细胞毒作用 ,10 -HCPT作用下T2 4人膀胱癌细胞DNA断裂损伤的增加 ,可能就是 10 Objective:To investigate the effect of 10-Hydroxyl camptothecine(10-HCPT)on DNA damage in T24 human bladder cancer.Methods:T24 human bladder cancer were incubated with various doses(0 01~0 1mg/ml) of 10-HCPT for different time duration(24 h or 48h).DNA damage was measured by single-cell gel electrophoresis(SCGE)and laser scanning confocal microscopy(LSCM).The degree of DNA damage was judged by the percentage of bladder cancer with comet-like tail,the tail length and tail moment of the cell.Results:DNA damage was demonstrated dose dependently in T24 human bladder cancer when the cells were incubated with 10-HCPT for certain time.The percentage of T24 human bladder cancer with comet-like tail,tail length and tail moment were aggravated significantly( P <0 05, P <0 01).Conclusions:The 10-HCPT had great cytotoxic effect on bladder cells the results indicate that the increasing of DNA damage in T24 human bladder cancer may be very important mechanism of 10-HCPT anti-tumor effects.
