摘要
目的 探讨开展血浆去除白细胞工作的必要性。方法 用Nageotte计数法、流式细胞分析和双向混合淋巴细胞培养的方法 ,对血浆中白细胞的数量、亚群情况、存活情况及同种免疫原性进行分析。结果 新鲜血浆中残余的白细胞约为 3.3×10 5/袋 ,经过 3个月的冻存后尚有近 30 %的白细胞存活并显示一定的同种免疫活性。结论 新鲜血浆的输注有引起同种免疫作用的危险 ,大量使用新鲜血浆如血浆置换等必须考虑白细胞的去除 ,如经济条件允许 ,输注去白细胞的血浆无疑是最佳选择。
Objective To explore the importance of leukocytes elimination from plasma products.Methods The quantity of leukocytes in plasma products was estimated by Nageotte method.The viability and subsets of leukocytes were determined by flow cytometer.Double mixed lymphocyte cultures (MLC) were performed to investigate alloimmunization of residual leucocytes in plasma products. Results The average number of leukocytes in a single unit of blood was 3.3×105.After 3 months of freezing,about 30% of leukocytes were still alive and may bear alloimmune activity.Conclusion The risk of alloimmunization may be caused by transfusion of fresh plasma.The elimination of leukocytes must be seriously recognized in the patients who are receiving a large volume of fresh plasma,e.g.,plasma exchange therapy.
